Abstract Background Ecological imbalance of the gut microbiota may be an important endogenous trigger mastitis, but underlying mechanisms are unclear. Recent findings indicated that selected bacteria maternal gastrointestinal can reach mammary gland through entero-mammary pathway. ABSTRACT Healthy mammary glands are essential for high-quality milk production in the dairy industry. The relationship between somatic cell counts (SCCs), rumen fermentation, and microbiota interactions remains unclear. This study integrated physiological indicators, high-throughput 16S rRNA gene sequencing, metagenomics data analysis to investigate mechanisms linking microbes mastitis evaluate changes serum cytokine levels cows with low (L-SCC) high (H-SCC) counts. Compared L-SCC group, H-SCC group exhibited significantly lower lactose fat contents milk, reduced fermentation product levels, increased abundances of Bacteroidetes, Firmicutes, Lachnospiraceae , Prevotella Rumiclostridium. Elevated IgG2, IgM, IL-1β, IL-6, TNF-ɑ indicated inflammation dysbiosis. Functional microbial communities revealed significant enrichment pathways related glutathione metabolism, thyroid hormone synthesis, hypertrophic cardiomyopathy (HCM), phosphotransferase system (PTS), P53 signaling pathway, Jak-STAT pathway.
