Silk fibroin hydrogels occupy an essential position in the biomedical field due to their remarkable biological properties, excellent mechanical flexible processing as well abundant sources and low cost. Herein, we introduce unique structures physicochemical characteristics of silk fibroin, including biocompatibility, biodegradability. Then, various preparation strategies are summarized, which can be divided into physical cross-linking chemical cross-linking. Emphatically, applications hydrogel biomaterials fields, tissue engineering, drug delivery, wearable sensors, systematically summarized. At last, challenges future prospects discussed.

Protein-based therapeutics have revolutionized the pharmaceutical industry and become vital components in development of future therapeutics. They offer several advantages over traditional small molecule drugs, including high affinity, potency specificity, while demonstrating low toxicity minimal adverse effects. However, manufacturing processes protein-based presents challenges related to protein folding, purification, stability immunogenicity that should be addressed. These proteins, like other biological molecules, are prone chemical physical instabilities. Microneedles are a patient-friendly technique for delivering drugs to the site of action in place traditional oral and injectable administration. Silk fibroin represents an interesting polymeric biomaterial because its mechanical properties, thermal stability, biocompatibility possibility control via genetic engineering. This review focuses on critical research progress silk microneedles since their inception, analyzes detail structure properties fibroin, types microneedles, drug delivery applications clinical trials, summarizes future development trend this field. It also proposes direction including increasing loading doses enriching as well exploring with stimulation-responsive release functions. Covalent conjugation and non-covalent complexation strategies for intracellular delivery of proteins using cell-penetrating peptides DOI Open Access

Therapeutic proteins provided new opportunities for patients and high sales volumes. However, they are formulated extracellular targets. The lipophilic barrier of the plasma membrane renders vast array intracellular targets out reach. Peptide-based delivery systems, namely cell-penetrating peptides (CPPs), have few safety concerns, low immunogenicity, with control over administered doses. This study investigates CPP-based protein systems by classifying them into CPP-protein "covalent conjugation" CPP: "non-covalent complexation" categories. Covalent conjugates ensure proximity CPP to cargo, which can improve cellular uptake endosomal escape. We will discuss various aspects covalent through non-cleavable (stable) or cleavable bonds. Non-cleavable produced recombinant DNA technology express complete fusion in a host cell chemical ligation protein, ensures stability during process. bonds classified pH-sensitive redox-sensitive bonds, enzyme-cleavable physical stimuli linkers (light radiation, ultrasonic waves, thermo-responsive). highlighted key characteristics non-covalent complexes electrostatic hydrophobic interactions preserve conformational integrity cargo. CPP-mediated complexation, such as zippers, adaptor methods, avidin-biotin technology, featured. Conclusively, complexation methods appropriate when number samples be screened. In contrast, biological activity is critical compartment, conjugation protocols preferred.
