Background Delivery systems based on albumin nanoparticles (NPs) have recently garnered substantial interest in anti-tumor drug development. However, systematic bibliometric analyses this field remain lacking. This study aimed to analyze the current research status, hotspots, and frontiers application of NPs oncology from a perspective. Methods Using Web Science Core Collection (WOSCC) as data source, retrieved articles were analyzed using software, such VOSviewer 1.6.18 CiteSpace 6.1.6, relevant visualization maps plotted. Cancer is among the leading causes of death worldwide. Therefore, timely diagnosis and appropriate treatment are very important. There many disadvantages that come with traditional cancer treatments, such as chemotherapy radiotherapy. In these specific drug concentration affects not only tumor site but also healthy tissues or organs. One foremost promising uses nanotechnology in field medical technology targeting can be achieved due to manipulation materials at a nanometric scale. Nanotechnology offers benefits terms therapy by enhancing it reducing its adverse effects guiding drugs selectively target cells. addition, use minute amounts medicine lead cost savings. Furthermore, nanoparticles used imaging agents improve diagnostics, therapeutics, management. Thus, this review has focused on different types therapy, their action mechanisms, applications diagnosis, imaging, treatment. Abstract Background The general sluggish clearance kinetics of functional inorganic nanoparticles tend to raise potential biosafety concerns for in vivo applications. Renal is a possible elimination pathway delivered through intravenous injection, but largely depending on the surface physical chemical properties given particle apart from its size and shape. Results In this study, three small-molecule ligands that bear diphosphonate (DP) group, different terminal groups other side, i.e. , anionic, cationic, zwitterionic groups, were synthesized used modify ultrasmall Fe 3 O 4 evaluating structure-dependent renal behaviors. Systematic studies suggested variation did not significantly increase hydrodynamic diameter nanoparticles, nor influence their magnetic resonance imaging (MRI) contrast enhancement effects. Among samples, nanoparticle coated with ligands, @DMSA, exhibited optimal efficiency reduced reticuloendothelial uptake. Therefore, sample was further labeled 99m Tc DP moieties achieve renal-clearable MRI/single-photon emission computed tomography (SPECT) dual-modality nanoprobe. resulting nanoprobe showed satisfactory capacities 4T1 xenograft tumor mouse model. Furthermore, biocompatibility @DMSA evaluated both vitro safety assessment experiments. Although cancer immunotherapy has made great strides in the clinic, it is still hindered by tumor immunosuppressive microenvironment (TIME). The stimulator of interferon genes (STING) pathway which can modulate TIME effectively emerged as a promising therapeutic recently. However, delivery most STING agonists, specifically cyclic dinucleotides (CDNs), performed intratumorally due to their insufficient pharmacological properties, such weak permeability across cell membranes and vulnerability nuclease degradation. To expand clinical applicability CDNs, novel pH-sensitive polycationic polymer-modified lipid nanoparticle (LNP-B) system was developed for intravenous CDNs. LNP-B significantly extended circulation CDNs enhanced accumulation within tumor, spleen, tumor-draining lymph nodes compared with free thereby triggering dendritic cells repolarizing pro-tumor macrophages. These events subsequently gave rise potent anti-tumor immune reactions substantial inhibition tumors CT26 colon cancer-bearing mouse models. In addition, acid-sensitive property polymer, more biocompatible safer nanoparticles formulated an indissociable cationic DOTAP (LNP-D). findings suggest that potential immunotherapy.
