Abstract Macrophages are immune cells belonging to the mononuclear phagocyte system. They play crucial roles in defense, surveillance, and homeostasis. This review systematically discusses types of hematopoietic progenitors that give rise macrophages, including primitive progenitors, erythro-myeloid stem cells. These have distinct genetic backgrounds developmental processes. Accordingly, macrophages exhibit complex diverse functions body, phagocytosis clearance cellular debris, antigen presentation, response, regulation inflammation cytokine production, tissue remodeling repair, multi-level regulatory signaling pathways/crosstalk involved homeostasis physiology. In this report, we detail the discovery and characterization of a new oligosaccharide, PCO-1, extracted from culture broth marine fungus Penicillium citrinum SCAU-268. The molecular weight PCO-1 was accurately measured to be 1476 Da, its primary structure identified as α-D-Glcp-(1,4)-α-D-Glcp-(1,4)-α-D-Glcp-(1,6)-α-D-Glcp-(1,6)-α-D-Glcp-(1,4)-α-D-Glcp-(1,4)-α-D-Glcp-(1,4)-α-D-Glcp-(1,4)-α-D-Glcp. Our results demonstrate that markedly elevated secretion cytokines IL-6, IL-1β, TNF-α, nitric oxide (NO) in concentration-dependent fashion. Metabolomic studies suggest may act through LACC1 pathway, promoting conversion citrulline ornithine modulating immune responses. conclusion, our study has successfully elucidated fungal-derived oligosaccharide confirmed potent immunomodulatory activity.

Photodynamic therapy (PDT)-based photoimmunotherapy has attracted increasing attention in the field of cancer immunotherapy. Nonetheless, monotherapy alone proves insufficient eliciting robust and enduring tumor immunogenicity within "cold" microenvironment triple-negative breast cancer. Therefore, it is imperative to integrate phototherapy immunostimulation strategies achieve synergistic effects. Here, we developed a carrier-free nanocombo comprising photosensitizer (chlorin e6, Ce6) an α2-adrenergic receptor (α2-AR) agonist (guanfacine, GFC) enhance through α2-AR activation. Ce6 GFC possessed ability self-assemble into spherical nanoparticles, with resulting Ce6-GFC (CeG) exhibiting exceptional drug loading efficiency (approaching 100%) long-lasting colloidal stability, along effective vivo tumor-targeting capabilities. Following near-infrared laser irradiation, CeG-mediated instigated rapid generation reactive oxygen species, leading membrane disruption release tumor-associated antigens, thereby facilitating dendritic cell maturation. Furthermore, agonists served repolarize M2 macrophages toward M1 phenotype via adenylyl cyclase-mediated activation α2-AR, promoting recruitment cytotoxic T lymphocytes. As result, significantly enhanced efficacy combatting poorly immunogenic tumors female mice. Our findings showcase "killing two birds one stone" approach that boosts immunogenicity, mitigates immunosuppression, advances photoimmunotherapy.
