Exosomes are nano-sized extracellular vesicles that play essential roles in intercellular communication, carrying biomolecules such as proteins, lipids, and RNAs can influence physiological pathological processes. The isolation of pure exosomes is critical for both basic research clinical applications, including diagnostics therapeutics. Traditional exosome techniques, ultracentrifugation, lack specificity may yield impure samples, making the need advanced techniques evident. Ligand-based affinity purification (LEAP) column chromatography has emerged a novel method utilizes specific ligands targeting surface markers, providing highly specific, gentle, scalable approach to isolation. This mini review explores LEAP chromatography’s mechanism, benefits, potential emphasizing its g.rowing importance exosome-based therapies.

ABSTRACT Background Exosomes, an emerging treatment of interest to aesthetic dermatology, have therapeutic applications in skin rejuvenation, alopecia, atopic dermatitis, acne scarring, and wound healing. Platelet‐rich plasma has been widely utilized for the same indications, among others. Currently, there are no trials comparing two regenerative modalities dermatology. Aims To compare efficacy safety adipose mesenchymal stem cell‐derived (ASC) exosomes versus platelet‐rich photoaged facial skin. Methods An investigator‐blinded, split‐face trial was conducted. Participants with mild moderate photoaging underwent three radiofrequency microneedling treatments PRP topical each applied one half face. Results Both equally improved wrinkling, dyschromia, erythema, texture, overall appearance. Histological analysis confirmed increased collagen I glycosaminoglycans, without significant differences between arms. Conclusion ASC a promising PRP‐alternative that may be attractive needle‐averse patients can hasten office visit duration, as phlebotomy centrifugation not required.

ABSTRACT Background There are exciting advances in the field of exosomes research as a potential regenerative therapy. Platelet‐rich plasma represents an abundant source exosomes. Aim To present evidence on use platelet‐rich plasma‐derived dermatology and discuss current technical limitations. Methods A literature search was conducted using Embase (Ovid), Web Science, ClinicalTrials.gov June 2024. Results contain myriad growth factors, genetic materials, lipids that mediate paracrine signaling. Several preclinical studies have demonstrated their wound healing. This includes faster contraction diabetic animal models, well enhanced angiogenesis, increased cell proliferation migration, protection against hyperglycemic conditions vitro. case study reported topical platelet‐derived exosome product tried successfully patient with persistent scalp wounds. In contrast, there is scarce information hair skin rejuvenation. Isolation techniques, activation methods, methods delivery not been optimized, which warrants further research. Novel such hydrogel‐based preparations, may enable application Conclusion Exosomes derived from therapeutic potential. More needed to standardize its use.
