Triphala is a traditional Ayurvedic herbal formulation composed of three fruits: amla (Phyllanthus emblica), bibhitaki (Terminalia bellerica), and haritaki chebula). potent remedy that promotes digestion, detoxification, overall wellness, while also providing antioxidant benefits through its trio nutrient-rich fruits. In order to elucidate the individual contributions ingredients from molecular perspective, were used for untargeted LCMS/MS analysis. Fresh fruits (PE, TC, TB) collected, processed into coarse powders, sequentially extracted {hexane, chloroform, ethyl acetate}. data analysis was performed on resultant metabolites, with bioinformatics tools employed pathway enrichment, target prediction, classification identified compounds. Additionally, polyphenols as key compounds potential health benefits. LCMS extracts total 10227 features, resulting in 2515 annotated PE contributing highest number at 1286. Comparative revealed 408 non-redundant 74.2% being unique fruits, underscoring complementary phytochemical profiles. Pathway enrichment highlighted dominant phenylpropanoid biosynthesis pathways across all extracts, comprehensive polyphenol 71 polyphenols, significant interactions predicted between gut microbiota. five common showed human targets related activity. These findings provide deeper understanding diversity Triphala, supporting use promoting health.

Cancer is one of the most concerning public health issues and breast cancer common cancers in world. The immune cells within tumor microenvironment regulate development. In this study, single cell data sets were used to identify marker gene for exhausted CD8 + T (CD8Tex) cancer. Machine learning methods cluster subtypes establish prognostic models with bulk using evaluate impacts CD8Tex. We analyzed overexpressing survival-associated genes identified CD8Tex hub protein-protein-interaction network. relevance T-cells was evaluated. clinical associations sequencing spatial data. pan-cancer expression, survival, association analyzed. biomarker CD8Tex-based subtyping systems performed well separation patients different survival. CRTAM, CLEC2D, KLRB1 as demonstrated have potential therapy impact. This study provides a unique view critical therapy.

1,3,6-Trigalloylglucose is a natural compound that can be extracted from the aqueous extracts of ripe fruit Terminalia chebula Retz, commonly known as “Haritaki”. The potential anti-Helicobacter pylori (HP) activity this has not been extensively studied or confirmed in scientific research. This was isolated using semi-preparative liquid chromatography (LC) system and identified through Ultra-high-performance chromatography–MS/MS (UPLC-MS/MS) Nuclear Magnetic Resonance (NMR). Its role evaluated Minimum inhibitory concentration (MIC) assay minimum bactericidal (MBC) assay, scanning electron microscope (SEM), inhibiting kinetics curves, urea fast test, Cell Counting Kit-8 (CCK-8) Western blot, Griess Reagent System. Results showed effectively inhibits growth HP strain ATCC 700392, damages structure, suppresses Cytotoxin-associated gene A (Cag A) protein, crucial factor infection. Importantly, it exhibits selective antimicrobial without impacting normal epithelial cells GES-1. In vitro studies have revealed acts an anti-adhesive agent, disrupting adhesion to host cells, critical step These findings underscore targeted therapeutic agent against infections.

Terminalia chebula Retz, known as the King of Tibet, is considered a functional food in China, celebrated for its antioxidant, immune-modulating, antibacterial, and anti-inflammatory properties. Chebulinic acid, derived from aqueous extracts However, potential an anti-Helicobacter pylori (HP) agent has not been fully explored. Herein, we extracted main compound Retz using semi-preparative liquid chromatography (LC) system identified 5 chebulinic acid through Ultra-high performance chromatography-MS/MS (UPLC-MS/MS) Nuclear Magnetic Resonance (NMR). To evaluate role, conducted minimum inhibitory concentration (MIC) bactericidal (MBC) assays, scanning electron microscope (SEM) imaging, inhibiting kinetics curves, urea fast test, cell counting kit-8 (CCK-8) assay, western blot analysis, griess reagent system, molecular docking. Our results showed that effectively inhibited growth HP strain ATCC 700392, damaged structure, exhibited selective antimicrobial activity without affecting normal epithelial cells GES-1. Importantly, it suppressed expression Cytotoxin-associated gene A (Cag A) protein, crucial factor infection. Molecular docking analysis predicted strong affinity (-9.7 kcal/mol) between Cag protein. Overall, our findings suggest acts anti-adhesive agent, disrupting adhesion to host cells, which critical step It also suppresses These highlight against infections.

Background Helicobacter pylori ( H. ) is thought to primarily colonize the human stomach and lead various gastrointestinal disorders, such as gastritis gastric cancer. Currently, main eradication treatment triple or quadruple therapy centered on antibiotics. Due antibiotic resistance, rate of decreasing gradually. Therefore, searching for anti- drugs from herbal sources has become a strategy treatment. Our team proposed Hezi Qingyou Formula (HZQYF), composed Chebulae Fructus, Ficus hirta Vahl Cloves, studied its activity mechanism. Methods Chemical components HZQYF were using UHPLC–MS/MS HPLC. Broth microdilution method agar dilution used evaluate HZQYF’s antibacterial activity. The effects expression adhesion genes alpA , alpB babA ), urease ureE ureF flagellar flaA, flaB explored Reverse Transcription-quantitative Polymerase Chain Reaction (RT-qPCR) technology. Effects morphology permeability extracellular membrane scanning electron microscopy (SEM) N-phenylnaphthalen-1-amine (NPN) uptake. Effect was kinetics analysis in vitro . Immunofluorescence staining examine effect adhesion. Western blot cagA protein. Results Minimum inhibitory concentration (MIC) values formula against clinical strains standard 80–160 μg/mL, minimum bactericidal (MBC) 160–320 μg/mL. could down-regulate alp A, flaA change increase permeability, decrease Conclusion Present studies confirmed that had promising activities demonstrated possible mechanism action by down-regulating bacterial adhesion, urease, gene expression, which provided scientific bases further investigations.

Background: Bacterial lysates, as immunomodulators, may play a role in regulating the host immune response. Objectives: This study aimed to investigate effects of bacterial lysates OM85-BV treating pulmonary infections caused by Streptococcus pneumoniae children. Methods: A total 120 children with S. infection were divided into two groups: The control group and group, 1: 1 ratio. received intravenous mezlocillin at 100 mL/kg/d, while addition mezlocillin, orally 3.5 mg per dose. improvement clinical symptoms function indexes—including respiratory rate, tidal volume, volume peak expiratory flow proportion exhaled time time—were compared. Additionally, levels C-reactive protein (CRP), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), soluble interleukin-2 receptor (sIL-2R), T lymphocyte subsets, immunoglobulins serum analyzed. Results: Compared showed significant decrease rate an increase flow, (P < 0.05 for all). also had significantly lower inflammatory factors such CRP, TNF-α, IL-6, sIL-2R. In terms function, increased subsets CD3+, CD4+, CD4+/CD8+ ratios, CD8+, well elevated IgA IgG levels. effective treatment was higher than 0.05). Conclusions: improves symptoms, lung reducing appears inhibit activation toll-like receptors (TLRs) nuclear factor kappa-B (NF-κB) pathway.
