A consensus single-cell transcriptomic atlas of dermal fibroblast heterogeneity

sequencing
(scRNAseq)
studies
have
unveiled
large
transcriptomic
heterogeneity
within
both
human
and
mouse
dermal
fibroblasts,
but
a
consensus
atlas
that
spans
species
is
lacking.
Here,
by
studying
25
9
datasets
through
semi-supervised
procedure,
we
categorize
15
distinct
fibroblast
populations
across
5
main
axes.
Analysis
of
markers
characteristic
each
population
suggested
diverse
functions,
such
as
position-dependent
ECM
synthesis,
association
with
immune
responses
or
structural
roles
in
skin
appendages.
Similarly,
fibroblasts
were
categorized
into
17
Comparison
highlighted
similarities
suggesting
degree
functional
overlap,
though
nuanced
differences
also
noted:
transcriptomically,
axes
seem
to
segregate
function,
while
prioritize
positional
information
over
function.
Importantly,
addition
newer
did
not
significantly
change
the
defined
structure.
This
study
enhances
our
understanding
diversity,
shedding
light
on
species-specific
distinctions
well
shared
functionalities.

Chronic
kidney
disease
affects
1
in
10
people
globally,
with
a
prevalence
twenty
times
that
of
cancer.
A
subset
individuals
will
progress
to
end-stage
renal
(ESRD)
where
replacement
therapy
is
required
maintain
health.
Cutaneous
disease,
including
xerosis
and
pruritus,
are
endemic
amongst
patients
ESRD.
In
the
uraemia-associated
immune
deficiency
ESRD,
impaired
circulating
responses
contribute
increased
infection
risk
poorer
vaccination
response.
Clinical
manifestations
