The Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is a betacoronavirus responsible for the COVID-19 pandemic, causing respiratory disorders, and even death in some individuals, if not appropriately treated time. To face preventive measures have been taken against contagions application of vaccines to prevent severe disease cases. For treatment, antiviral, antiparasitic, anticoagulant other drugs reused due limited specific medicaments disease. Drug repurposing an emerging strategy with therapies that already tested safe humans. The pandemic of coronavirus disease 2019 (COVID-19) has a broad spectrum clinical manifestations. severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) undergoes continuous evolution, resulting in the emergence several variants. Each variant different severity and mortality rate.In this study, 1174 COVID-19 patients were studied for over three SARS-CoV-2 predominating periods 2021 2022 Sulaimani Province, Iraq. In each period, representative, virus was subjected to phylogenetic molecular analysis.Phylogenetic analysis revealed variants, belonging to: Delta B.1.617.2, Omicron BA.1.17.2, BA.5.6. variants showed more symptoms lower PCR-Ct value than regardless gender, only 4.3% cases asymptomatic. rate with (.5% BA.5.2 1.3% BA.1.17.2) compared (2.5%). higher males (2.84%), while that BA1.17.2 females, 1.05% .0%, respectively. Age group (≥70) years had highest rate; however, it (.0%) age (30-49) (.96%) variants. There been surge infection city due predominant lineages SARS-CoV-2, B.1.617, BA.1.17.2 BA.5.6, A and/or significantly correlated death same order.

SARS-CoV-2 is an etiologic agent responsible for the coronavirus disease 2019 (COVID-19) pandemic. The virus has rapidly extended globally and taken millions of lives due to unavailability therapeutics candidates against virus. Till now, no specific drug have been developed that can prevent or treat infections caused by pathogen. main protease (Mpro) plays a pivotal role in mediating viral replication mechanistically inhibition protein hinder infection process Therefore, study aimed identify natural bioactive compounds block activity Mpro subsequently infections. Initially, total 96 phytochemicals from Ruellia prostrata Poir. Senna tora (L.) Roxb. plants were identified through gas chromatography-mass spectrometry (GC-MS) analytical method. Subsequently, screened molecular docking, absorption, distribution, metabolism, excretion (ADME), toxicity (T), dynamics (MD) simulation approach. docking method initially four molecules having PubChem CID: 70825, 25247358, 54685836 and, 1983 with binding affinity ranging between -6.067 -6.53 kcal mol-1 active site target protein. All selected exhibit good pharmacokinetics properties. Finally, further evaluated based on MD methods confirmed stability targeted computational approaches best be as treatment option disease-related complications. Although, experimental validation suggested evaluation work.

The ongoing coronavirus disease 2019 (COVID-19) pandemic represents one of the most exigent threats our lifetime to global public health and economy. As part pandemic, from January 10 March 10, 2020, severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) began spread in Hefei (Anhui Province, China) with a total 174 confirmed cases COVID-19. During this period, we were able gather critical information on transmission evolution pathogens through genomic surveillance. Particularly, objective study was track putative variants SARS-CoV-2 circulating for first time contribute effort toward elucidating molecular epidemic profile virus. Patients who showed symptoms COVID-19 routinely tested infections via RT-PCR at First Affiliated Hospital Anhui Medical University. Whole-genome sequencing performed 97 clinical samples collected 29 patients. result, identified local novel single-nucleotide polymorphism site (10,380) harboring G → T mutation (Gly Val) Hefei. Further phylogenetic network analysis all sequences deposited GenBank East Southeast Asia revealed subtype S-type (a1) C synonymous (Leu) position 18,060 ORF1b, likely representing that is obviously concentrated Yangtze River Delta region. Moreover, investigation inflammatory cytokine patients suggested mutations positions (the shared variable a1) 28,253(harboring mutation, Phe) associated milder immune responses

The global COVID-19 pandemic, caused by SARS-CoV-2, has created an urgent need for novel antiviral agents, as it led to a devastating loss of lives worldwide. In response, numerous vaccines were developed in 2022, with ongoing efforts more candidates. Despite significant progress, reports vaccine-related issues and side effects have emerged. effectiveness been challenged new viral variants, lack well-designed vivo experiments, randomised controlled trials. Currently, no clinically approved medications exist treating COVID-19. However, few broad-spectrum drugs shown promising outcomes clinical studies. Many scientists believe that natural compounds derived from herbal medicines mushrooms could offer substantial benefits patients. Ganoderma, known its traditional use promoting health longevity, rich history therapeutic applications dietary supplementation. Numerous epidemiological studies demonstrated Ganoderma derivatives can enhance innate adaptive immune responses against various pathogens, including viruses, laboratory settings living organisms. Therefore, the secondary metabolites such triterpenoids, polysaccharides, glycoproteins, hold potential developing drugs. These mechanisms, inhibition replication, modulation responses, direct virucidal effects. Given search effective treatments COVID-19, bioactive stand out candidates further research development. Their multifaceted mechanisms make them suitable exploration agents potentially contributing enhancing existing treatments.
