Objective To investigate the effectiveness of lacrimal gland ultrasonography in assessment chronic ocular graft-versus-host-disease (oGVHD) after allogeneic hematopoietic stem cell transplantation (allo-HSCT) and to establish correlation between surface ultrasonographic results. Method The cross-sectional study included 57 participants aged 18 older, who were at least 100 days allo-HSCT. was conducted oGVHD clinic Peking University People’s Hospital March June 2023. Patients categorized into groups according International Chronic (ICCGVHD) consensus group diagnostic criteria or 2005 National Institutes Health (NIH) classification for GVHD. Demographics transplantation-related information collected all participants, including age, gender, donor-recipient HLA matching, ABO gender combination duration disease activity severity involvement assessed using various parameters such as Ocular Surface Disease Index (OSDI), Schirmer test, tear film break-up time (BUT), meniscus height, corneal/conjunctival staining meibomian dropout. Lacrimal structures by B-mode Doppler measure long diameter, thick homogeneity parenchymal vascularization. Statistical analyses performed determine differences conditions well condition findings. Result (1) with definite probable exhibited a significantly longer allo-HSCT compared non-oGVHD patients (H=11.264, p<0.01), median durations 247(164,894) 525(310,928) days, respectively, 204(169,323.25) patients. (2) Compared patients, both showed lower average test (H=31.188, TBUT (H=11.853, height (H=13.630, p<0.01) higher OSDI (F=27.992, corneal scores (χ²=23.66, p<0.05) temporal conjunctival (χ²=14.84, p<0.05). (3) glands vascularization did not exhibit significant three groups. (4) diameter had positive correlations (r=0.297, negative (r=-0.313, total (r=-0.285, demonstrated (r=0.404, (r=-0.273, p<0.05), (r=-0.264, nasal (r=-0.271, (r=-0.312, Homogeneity found be correlated status. Purpose: Aging alters the function of lacrimal gland and disrupts balance microenvironment on ocular surface, eventually leading to aqueous-tear-deficient dry eye. Mitophagy has been reported play an important role in aging, but underlying mechanism remains unclear. Methods: The young (6 weeks) middle-aged (12 months) male C57BL/6J mice were used this study, mitophagy agonist rapamycin inhibitor Mdivi-1 vivo experiments. Hematoxylin eosin, Masson, Oil Red O, reactive oxygen species (ROS) staining detect histological changes lipids gland. Changes expression proteins identified by Western blotting lysates. Transmission electron microscopy immunofluorescence assess mitophagy. single-cell RNA sequencing (scRNA-seq) bioinformatics analyses transcription signature during aging.
