Abstract The angiotensin-converting enzyme 2 (ACE2) is a primary cell surface viral binding receptor for SARS-CoV-2, so finding new regulatory molecules to modulate ACE2 expression levels promising strategy against COVID-19. In the current study, we utilized islet organoids derived from human embryonic stem cells (hESCs), animal models and COVID-19 patients discover that fibroblast growth factor 7 (FGF7) enhances within islets, facilitating SARS-CoV-2 infection resulting in impaired insulin secretion. Using hESC-derived organoids, demonstrated FGF7 interacts with FGF (FGFR2) FGFR1 upregulate predominantly β cells. This upregulation increases both secretion susceptibility of infection. Inhibiting FGFR counteracts FGF7-induced upregulation, subsequently reducing replication islets. Furthermore, retrospective clinical data revealed diabetic severe symptoms exhibited elevated serum compared those mild symptoms. Background. Coronavirus disease 2019 (COVID-19), caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, has led to widespread illness and global health challenges. While phase of infection been extensively studied, long-term effects, particularly in individuals who have recovered, remain unclear. Post-acute complications SARS-CoV-2 involves prolonged symptoms systemic disorders, including persistent inflammation, endothelial dysfunction, immune dysregulation, which often last for months. Among key factors contributing these conditions, there are pro-inflammatory cytokines, growth involved angiogenesis, hypoxia-inducible stress-related proteins, play a significant role tissue repair response modulation.
