Radiotherapy (RT) is a primary clinical approach for cancer treatment, but its efficacy often hindered by various challenges, especially radiation resistance, which greatly compromises the therapeutic effectiveness of RT. Mitochondria, central to cellular energy metabolism and regulation cell death, play critical role in mechanisms radioresistance. In this context, cuproptosis, novel copper-induced mitochondria-respiratory-dependent death pathway, offers promising avenue radiosensitization. study, an innovative theranostic nanoplatform was designed induce cuproptosis synergy with low-dose therapy (LDRT, i.e., 0.5–2 Gy) treatment situ hepatocellular carcinoma (HCC). This aims reverse hypoxic tumor microenvironment, promoting shift from glycolysis oxidative phosphorylation (OXPHOS), thereby enhancing sensitivity cuproptosis. Concurrently, Fenton-like reaction ensures sustained supply copper depletion glutathione (GSH), inducing disrupting mitochondrial function, interrupting supply. strategy effectively overcomes radioresistance enhances against tumors. conclusion, study elucidates intricate interactions among hypoxia reversal, metabolic reprogramming, radiosensitization, particularly context treating carcinoma, providing paradigm radiotherapy.

High invasiveness mesothelioma is a malignant tumor of the peritoneum or pleura. The effect cuproptosis on (MESO) still unknown, though. Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) datasets were used to identify differential genes linked in mesothelioma. Multigene features then created assess course disease. Use single-cell data vitro validation uncover crucial gene regulation mechanisms. In MESO, we found nine differentially expressed cuproptosis. Using univariate Cox LASSO regression techniques, 3-gene feature (P < 0.05) was created, showing good predictive potential for survival time. According risk score, patients low-risk subset had considerably greater rate than those high-risk = 0). similar pattern prediction performance are also seen queue. findings drug sensitivity research indicate that patients, vinblastine, paclitaxel, gefitinib, erlotinib sensitive medications 0.05). Classical monocytes identified as core cells connected by CellChat results. SLC31A1 implicated positive M2 macrophage polarization, according cell subtype analysis confirmation. Genes have major influence immunity can predict how MESO will progress.

Copper is an essential micronutrient involved in various physiological processes cell types. Consequently, dysregulation of copper homeostasis-either excessive or deficient-can lead to pathological changes, such as heart failure (HF). Recently, a new type copper-dependent death known cuproptosis has drawn increasing attention the impact dyshomeostasis on HF. Notably, was associated with occurrence Hence, this review aimed investigate biological uptake, transport, excretion, and storage at both cellular systemic levels terms HF, along underlying mechanisms action. Additionally, role its related mitochondrial dysfunction HF pathogenesis analyzed. Finally, we reviewed therapeutic potential current drugs that target metabolism for treating Overall, conclusions revealed copper-based therapies development strategies treatment

A total 240 35-day-old rabbits were randomly allocated to five treatments (n = 6 per treatment, each replicate consisted eight rabbits) that included control group (CON) a basal diet, SAL 60 mg/kg salinomycin (SAL) in addition NCCC I, II, III groups, which supplemented 50, 100, 200 NCCC, respectively, diet. The test lasted for 28 d. results showed increased liver Cu/Zn-SOD content up-regulated gene expression (p < 0.05), while also reducing MDA enhancing antioxidant capacity Moreover, diet reduced IL-6 down-regulated relative IL-1β genes jejunum 0.05). Cuproptosis is a novel cell death dependent on mitochondrial respiration and regulated by copper. While the study of it mainly focused tumor therapy, in present study, two key cuproptosis-related genes, ferredoxin (FDX1) dihydrolipoamide S-acetyltransferase (DLAT) homologs (designated as CgFDX1 CgDLAT), were identified from Crassostrea gigas. has Fer2 domain with 2Fe-2S cluster forming unique ferredoxin. CgDLAT composed biotin_lipoyl domain, an E3-binding 2-oxoacid_dh domain. mRNA expressed all examined tissues. After elesclomol treatment, both protein expressions them reduced hemocytes. The mortality rate hemocytes increased significantly, accompanied noticeable adhesive abnormalities heightened secretion after treatment. Additionally, accumulation or depletion actin was observed integrity double membrane structure mitochondria compromised, organization cristae disrupted. contents copper, malondialdehyde (MDA), pyruvic acid mitoSOX well ratio cells low potential significantly upon treatment content citric decreased significantly. These findings suggest presence cuproptosis oysters its activation mechanism relatively conserved evolution.
