Adipose-derived regenerative cells (ADRCs) also referred to as the stromal vascular fraction, provide an ample source of stem with widespread therapeutic use. Being heterogenous in nature, possibly affecting clinical outcome after cell treatment, ADRC- donor, -BMI, and -gender may have a large impact on ADRC composition quality but this remains largely unexplored. Herein, we comprehensive single-cell RNA sequencing mapping across two trial intervention studies found no gender- or BMI-related variations, except for minor female increase PI16/CD55-expressing cells. Indeed, heterogeneity was surprisingly minimal between donors. This provides important decision-making support adipose donor selection treatments, suggest that gender BMI should be regarded less influential.

Abstract Adipose-derived stem cells (ASCs) provide an ample, easily accessible source of multipotent cells, alternative to bone marrow-derived stromal (BMSCs), capable differentiating into osteoblasts. However, the osteogenic potential ASCs is reportedly lower than that BMSCs and protocols effectively differentiate osteoblasts are in high demand. Injection of autologous adipose-derived regenerative cells (ADRCs) combined with lipotransfer has been suggested to alleviate symptoms in diseases including breast cancer-related lymphedema (BCRL). We recently performed a randomized controlled trial injecting lipoaspirate ADRCs into the axilla BCRL patients, and here we aimed intervention group define an unbiased fashion whether ADRC injection was linked clinical outcome. 39 patients received assisted (4.20 × 107 ± 1.75 cells) whereas 41 were included for placebo treatment. Mesenchymal stem cells (MSCs) have been proposed to treat osteoarthritis (OA) for many years. However, clinical outcomes inconsistent due biological variation between patients, differences in tissue source and preparation of the MSCs, type donor (e.g. allogenic versus autologous). Here, we test hypothesis that are related variations stemness senescence injected autologous adipose-derived (AD) MSCs. In prospective randomized trial, 45 knee OA patients were divided into two groups: Group 1 (n = 22) treated with high tibial osteotomy (HTO) alone 2 23) HTO followed by intra-articular injection AD-MSCs (HTO + AD-MSCs). MRI X-ray performed pre-operation 12 months post-operation. WOMAC VAS score collected four times, every 6 over a 24-month follow-up.
