Abstract Artificial intelligence (AI)-based multi-modal fusion algorithms are pivotal in emulating clinical practice by integrating data from diverse sources. However, most of the existing models focus on designing new modal methods, ignoring critical role feature representation. Enhancing representativeness can address noise caused heterogeneity at source, enabling high performance even with small datasets and simple architectures. Here, we introduce DeepOmix-FLEX (Fusion Learning Enhanced representation for X-modal or FLEX short), a model that integrates data, proteomic metabolomic pathology images across different scales modalities, advanced learning contains Feature Encoding Trainer structure train encoding, thus achieving inter-feature inter-modal. achieves mean AUC 0.887 prediction chronic kidney disease progression an internal dataset, exceeding 0.727 using conventional variables. Following external validation interpretability analyses, our demonstrated favorable generalizability validity, as well ability to exploit markers. In summary, highlights potential AI integrate optimize allocation healthcare resources through accurate prediction.

Here, we investigated different a series thiazolidinediones, including rosiglitazone, pioglitazone, second-generation compound MSDC-0602 K, on human mesenchymal stem cells (MSCs).MethodsWe developed 13C subcellular metabolomic tracer measuring separate mitochondrial cytosolic metabolite pools, lipidomic network-based isotopologue models, bioorthogonal chemistry, to demonstrate that K affected marrow-derived MSCs (BM-MSCs) adipose tissue-derived (AT-MSCs). In BM-MSCs, promoted osteoblastic differentiation suppressed adipogenesis. This effect was clearly from earlier drugs AT-MSCs.ResultsFluxomic data reveal unexpected differences between this drug's provide mechanistic insight into pharmacologic inhibition carrier 1 (MPC). Our study demonstrates retains capacity inhibit MPC, akin rosiglitazone but unlike enabling utilization alternative pathways. Notably, exhibits limited lipogenic potential compared both each which employs strategy.ConclusionsThese findings indicate new-generation do not compromise structure, offering safer for treating insulin resistance. Abstract According to traditional Chinese medicine (TCM) constitutional theory, individuals with phlegm-dampness constitution (PDC) are at increased risk for metabolic disorders. Previous studies have indicated that PDC exhibit gene expression changes associated disorders, even normal indices. However, the biological mechanisms underlying these remain unclear. The gut microbiota has recently emerged as a promising avenue elucidating TCM principles. Here, we revealed distinct and serum metabolite profiles. A decrease in phytosphingosine was scores disorder severity. Subsequent experiments demonstrated Flavonifractor plautii can biosynthesize phytosphingosine, which also negatively correlated score. Interestingly, both F. levels decreased subjects Fecal transplantation from accelerated development of disorders mice. supplementation ameliorated by increasing gut‒hepatic axis. Mechanistic investigations confirmed directly bind hepatic peroxisome proliferator-activated receptor α (PPARα) activate its nuclear transcription activity, thereby regulating downstream related glucose‒lipid metabolism. Our research indicates product, contributes increases their susceptibility These findings suggest diagnosing may be beneficial identifying at-risk populations among apparently healthy individuals, advancing broader field prevention integration.
