Multidrug efflux pumps are critical elements in both intrinsic and acquired antibiotic resistance of bacterial populations. Consequently, most studies regarding these protein machineries focus on this specific phenotype. Nevertheless, different works show that participate other aspects physiology too. Herein, we study the Pseudomonas aeruginosa multidrug pump MexJK. Previous studies, using model strains lacking MexAB-OprM MexCD-OprJ pumps, support MexJK can extrude erythromycin, tetracycline, triclosan. However, results here reported indicate potential increased extrusion, a mutant overexpressing mexJK, does not alter antibiotics susceptibility wild-type genetic background where all remain functional. clear impact quorum sensing (QS) response, mainly Pqs-dependent QS regulation network expression Pqs-regulated virulence factors, was observed linked to mexJK overexpression. The production siderophore pyoverdine strongly depended level expression, suggesting might P. pyoverdine-dependent iron homeostasis. All all, presented current article functions as MexJK, go beyond modulate relevant physiology.

Pseudomonas aeruginosa , a Gram-negative bacterium, is recognized for its adaptability and opportunistic nature. It poses substantial challenge in clinical settings due to complicated antibiotic resistance mechanisms, biofilm formation, capacity persistent infections both animal human hosts. Recent studies revealed potential zoonotic transmission of P. between animals, the environment, populations which highlights awareness this microbe. Implementation One Health approach, underscores connection human, animal, environmental health, we aim offer comprehensive perspective on current landscape management. This review presents innovative strategies designed counteract infections. Traditional antibiotics, while effective many cases, are increasingly compromised by development multidrug-resistant strains. Non-antibiotic avenues, such as quorum sensing inhibition, phage therapy, nanoparticle-based treatments, emerging promising alternatives. However, their application encounters obstacles like cost, side effects, safety concerns. Effectively addressing necessitates research efforts, advancements development, comprehension host-pathogen interactions deal with resilient pathogen.

Abstract Mutations in mexZ , encoding a negative regulator of the expression mexXY efflux pump genes, are frequently acquired by Pseudomonas aeruginosa at early stages lung infection. Although traditionally related to resistance first-line drug tobramycin, mutations associated with low-level aminoglycoside when determined laboratory, suggesting that their selection during infection may not be necessarily, or only, tobramycin therapy. Here, we show -mutated bacteria tend accumulate inside epithelial barrier human airway model, thus colonising epithelium while being protected against diverse antibiotics. This phenotype is mediated overexpression lecA quorum sensing-controlled gene, lectin involved P. tissue invasiveness. We find caused disrupted equilibrium between overproduced MexXY and another pump, MexAB, which extrudes sensing signals. Our results indicate affect sensing-regulated pathways, promoting invasiveness protecting from action antibiotics within patients, something unnoticeable using standard laboratory tests.

Collateral sensitivity is an evolutionary trade-off whereby acquisition of the adaptive phenotype resistance to antibiotic leads nonadaptive increased susceptibility another. The feasibility harnessing such a design evolutionary-based approaches for treating bacterial infections has been studied using model strains. However, clinical application collateral requires its conservation among strains presenting different mutational backgrounds. Particularly relevant studying robustness already-antibiotic-resistant mutants when challenged with new antimicrobial, common situation in clinics that hardly addressed. We submitted set diverse Pseudomonas aeruginosa antibiotic-resistant short-term evolution presence antimicrobials. Ciprofloxacin selects clinically mutations preexisting resistant mutants, which gave rise same, robust, aztreonam and tobramycin. then experimentally determined alternation ciprofloxacin more efficient than ciprofloxacin–tobramycin driving extinction analyzed mutants. Also, we show combinations ciprofloxacin–aztreonam or are most effective strategies eliminating tested P. These findings support identification conserved patterns may guide evolution-based treat infections, including those due Besides, this example phenotypic convergence absence parallel that, beyond antibiotic-resistance field, could facilitate understanding processes, where selective forces giving new, not clearly phenotypes remain unclear.

Bivalve shellfish consumption (ark shells, clams, cockles, and oysters) has increased over the last decades. Predicting evolution toward antibiotic resistance (AR) and its associated trade-offs, such as collateral sensitivity, is important to design evolution-based strategies tackle AR. However, the effect of nutrients' availability on evolution, particularly those that can be found under in vivo infection conditions, has been barely addressed.
