Due to the possible coexistence of Klebsiella pneumoniae (KP) and Candida albicans (CA), strains KP CA with biofilm production properties clinically isolated from patients were tested. The biofilms combined organisms (KP+CA) was higher than each organism alone, as indicated by crystal violet z-stack immunofluorescence. In parallel, bacterial abundance in + similar KP, but fungal (culture method), implying that grows better presence KP. Proteomic analysis performed compare alone. With mouse neutrophils (thioglycolate induction), induced less prominent responses biofilms, determined (i) neutrophilic supernatant cytokines (ELISA) (ii) neutrophil extracellular traps (NETs), using immunofluorescent images (neutrophil elastase, myeloperoxidase, citrullinated histone 3), peptidyl arginine deiminase 4 (PAD4) expression, cell-free DNA. Likewise, intratracheal C57BL/6 mice induces severe pneumonia organ injury (serum creatinine alanine transaminase) (colorimetric assays), (ELISA), bronchoalveolar lavage fluid parameters (bacterial culture abundances a hemocytometer), histology score (H&E stains), NETs (immunofluorescence on lung tissue). conclusion, biomass mostly produced potent activation Hence, fungi respiratory tract might benefit host some situations, despite well-known adverse effects fungi.

Abstract Aspiration pneumonia is a serious respiratory condition, which particularly prevalent in patients with dysphagia, neurological disorders, or those undergoing surgical interventions. The formation of multispecies biofilms the oral cavity, involving bacterial periodontopathogen Porphyromonas gingivalis and opportunistic pathogenic fungus Candida albicans , may also be related to development this disease, contributing resistance standard antimicrobial treatment. Therefore, research aimed evaluate efficacy selected antibiotics‒levofloxacin, metronidazole, meropenem, vancomycin‒and antifungal agents‒amphotericin B, caspofungin, fluconazole‒on these mixed aspiration model. While metronidazole levofloxacin effectively inhibited viability biofilms, lower doses increased release proteases. In conditions meropenem vancomycin showed reduced efficacy, requiring significantly higher achieve similar effect as single cultures. Treatment antifungals revealed that amphotericin B impacted fungal cell within was enhanced when drug applied presence P. . Caspofungin fluconazole variable caspofungin being more effective against C. cells biofilm. These findings indicated due mutual microbial protection mixed-species biofilm, retained its virulence despite increasing antibiotic doses. However, no excessive benefit observed for antifungals, indicating minor importance yeasts their protective role other pathogens infection. Graphical abstract

Siderophores are specialized molecules produced by bacteria and fungi to scavenge iron, a crucial nutrient for growth metabolism. Catecholate-type siderophores mainly bacteria, while hydroxamates mostly from fungi. This study investigates the capacity of nine hydroxamate-type Streptomyces facilitate iron acquisition human pathogen Pseudomonas aeruginosa. Growth assays under limitation 55Fe incorporation tests showed that all promoted bacterial transport. The also aimed identify TonB-dependent transporters (TBDTs) involved in import these siderophores. Using mutant strains lacking specific TBDT genes, it was found is imported into P. aeruginosa cells FpvB coprogen, triacetylfusarinine, fusigen, ferrirhodin, ferrirubin. Iron complexed desferioxamine G transported FoxA, ferricrocin-Fe ferrichrycin-Fe FiuA, rhodotoluric acid-Fe FpvB, another unidentified TBDT. These findings highlight effectiveness transport provide insights complex molecular mechanisms involved, which important understanding microbial interactions ecological balance.

Up to one-quarter of the United States population is affected by tinea pedis (athlete's foot). Tinea web space (interdigital pedis) a common clinical presentation causing skin macerations and fissures. A "dermatophytosis complex" (i.e., concomitant bacterial colonization) further complicates treatment. Here, we examined records 14,429 specimens taken from feet dermatology podiatry outpatients over 4.6-year period; all were subjected multiplex qPCR diagnosis for detection dermatophytes, Candida, Corynebacterium minutissimum, Pseudomonas Staphylococcus aureus. literature search was conducted review reported prevalence fungal agents. In both interdigital plantar foot specimens, dermatophytes (33.3-33.8%) S. aureus (24.3-25%) found be predominate pathogens. space, higher C. minutissimum (15.7% vs. 7.9%) (23.5% 9.6%) found. The more likely observed in presence reflecting risk mixed infection. dermatophyte-positive variant at 45.5% (SD: 2.3). An analysis patient characteristics showed male patients exhibiting likelihoods dermatophyte, detections. elderly disproportionately infected with Candida. children, an common, which could attributed impetigo. recent lacks reporting on colonization patients, due reliance culture supplemented antibiotics. Geographical variation has been identified Trichophyton mentagrophytes complex. conclusion, PCR serves as valuable tool management pedis. accurate timely pathogens can better inform healthcare providers appropriate treatment selection.

Historical processes in community assembly, such as species arrival order, influence interactions, causing priority effects. Candida albicans and Pseudomonas aeruginosa often co-occur biofilm-based infections of the skin, lungs, medical devices. Their predominantly antagonistic relationship involves complex physical chemical interactions. However, presence implications effects among these microorganisms remain largely unexplored. Here, we investigate impact effect dual-species biofilms using clinical isolates. By varying inoculation observe significant changes biofilm composition, structure, virulence, antimicrobial susceptibility. The first colonizer has an advantage for surface colonization. Consecutive colonization increases virulence negates C. albicans' protective on P. PAET1 against meropenem treatment. Background: Nontuberculous mycobacterial pulmonary disease (NTM-PD) is an increasingly prevalent chronic infection, where the host immune status plays a crucial role in susceptibility and progression. The complex microenvironment, characterized by diverse microbial communities interactions, exhibits distinct features that may be fundamentally altered patient’s underlying state. Methods: A total of 111 sputum specimens 64 bronchoalveolar lavage fluid (BALF) were collected from 143 patients diagnosed with NTM-PD under different states. Metagenomic sequencing was performed on these to characterize compare microenvironmental among through comprehensive bioinformatic analyses. Results: immunosuppressed group exhibited lower α-diversity (p < 0.05). Principal Coordinates Analysis (PCoA) β-diversity for BALF revealed significant differences between groups Linear discriminant analysis Effect Size (LEfSe) identified species enriched group. co-occurrence network indicated had more structured actively connected networks compared control Mantel test confirmed abundance associated clinical immune–inflammation-related indicators patients. Conclusions: Our study reveals microenvironment NTM-PD. Further work required explore two-way relationship micro-organisms inflammatory responses, influence patient outcomes.
