Abstract The utility of bicontinuous microemulsions (BMEs) as carriers the antimicrobial peptide (AMP) gramicidin D and antiseptic chlorhexidine was investigated for possible topical delivery to chronic wounds. two water‐insoluble solutes dissolved in pre‐formed one‐phase BMEs Water/Polysorbate 80/Limonene/Ethanol/Glycerol Water/Aerosol‐OT (AOT)/Polysorbate 85/Isopropyl Myristate an AOT/Polysorbate 85 Winsor‐III system, achieving concentrations 1.0 wt% 0.5% individually 0.3% mixtures at 22°C, respectively. Small‐angle neutron scattering measurements demonstrated that both decreased surfactant interfacial activity increased fluidity Polysorbate 80 system. For systems, ellipsoidal aggregates consisting likely adsorbed oil formed, while enhanced surface surfactants. According bioassays performed on artificial skin, incorporation melittin, gramicidin, general bioactivity 24 h treatment against relevant antibiotic‐resistant bacteria found skin relative controls. Yet, BME treatments were less effective than aqueous melittin control, contrast well diffusion previously. results reflect strong impact AMPs antiseptics structure dynamics complexity formulating optimal activity.

Abstract Recurrent urinary tract infections (UTIs) are increasingly associated with antibiotic resistance, posing significant clinical challenges. This study evaluates the antimicrobial efficacy of membrane-intercalating conjugated oligoelectrolyte COE-D8 against uropathogenic Escherichia coli (UPEC). Among 93 UPEC isolates, over 50% demonstrated resistance to first-line antibiotics, compound sulfamethoxazole showing highest (75% at a minimum inhibitory concentration (MIC) >512 μg/mL). significantly reduced rates, achieving less than 32 μg/mL and below 5% 512 μg/mL, while demonstrating least fourfold faster bacterial elimination standard antibiotics. Mechanistic studies revealed that disrupts membranes, inducing rapid lysis. However, its cytotoxicity in mammalian cells 2 limits application. To address this, combination therapy fosfomycin was explored, reducing toxicity by vitro. In mouse models, co-administration mitigated COE-D8-induced prevented mortality higher doses without compromising efficacy. These findings highlight potential COE-D8, particularly fosfomycin, as promising strategy combat antibiotic-resistant addressing

Abstract Despite significant advances in recent decades, severe burns remain a formidable challenge, with high morbidity and mortality rates. The immunocompromised state following burn injuries, compounded by the loss of protective skin barrier, increases risk bacterial colonization invasion. Without appropriate management, infections patients can progress to sepsis, life‐threatening complication. Current care often fails achieve optimal tissue regeneration infection prevention, necessitating combination different approaches. Developing innovative safer strategies mitigate is essential for improving patient outcomes. This review provides updated insights into various biomaterials tailored managing burns, offering comprehensive summary emerging technologies potential clinical application. Additionally, an in‐depth discussion on current research areas that warrant further investigation presented. Potential avenues next‐generation dermal substitutes aimed at preventing wound are then explored.
