Sinisan (SNS) consists of four kinds herbs, which is the core antidepressant prescription widely used in traditional Chinese medicine clinic treatment for depression induced by early life stress. Mitochondria are essential for energy production and reactive oxygen species (ROS) generation, with changes in ROS levels or demands affecting mitochondrial DNA (mtDNA) copy numbers, indicating function. Early life adversity (ELA) affects dynamics, influencing long-term health. Both ELA abnormalities have been independently associated bipolar disorder (BD). This study aims to explore the complex interplay between dysfunction, ELA, BD. The included 60 participants diagnosed BD 66 healthy controls (HCs). Data were collected using Childhood Trauma Questionnaire (CTQ), leukocyte mtDNA number (MCN) was determined from blood samples. results indicated CTQ sum scores significantly higher group, reflecting greater exposure ELA. In HCs, a marginally significant nonlinear relationship square of score MCN found. Further analysis demonstrated interaction on (p = 0.023), highlighting critical connection dysfunction reinforcing its biological underpinnings. Preclinical evidence suggests that antidepressants (ADs) may differentially influence mitochondrial energetics. This study was conducted to investigate the relationship between function and illness vulnerability in bipolar disorder (BD), specifically risk of treatment-emergent mania (TEM). Participants with BD already clinically phenotyped as TEM+ (n = 176) or TEM- 516) were further classified whether TEM associated AD, based on preclinical studies, increased (Mito+, n 600) decreased (Mito-, 289) electron transport chain (ETC) activity.
