The translocator protein (TSPO) positron emission tomography (PET) can noninvasively detect neuroinflammation associated with epileptogenesis and epilepsy. This study explored the role of TSPO-targeting radioligand [18F]F-TFQC, an m-trifluoromethyl ER176 analog, in PET neuroimaging epileptic rats. Initially, [18F]F-TFQC was synthesized a radiochemical yield 8%–10% (EOS), purity over 99%, specific activity 38.21 ± 1.73 MBq/nmol (EOS). After determining that exhibited good biochemical properties, performed rats at multiple time points various stages disease progression. imaging showed uptake right hippocampus (KA-injected site, i.e., epileptogenic zone), which most pronounced 1 week (T/NT 1.63 0.21) month 1.66 0.20). results were further validated using autoradiography pathological analysis. Thus, reflect TSPO levels localize zone, thereby offering potential for monitoring guiding anti-inflammatory treatment patients

Temporal lobe epilepsy (TLE) is associated with abnormal dynamic functional connectivity patterns, but the changes in brain activity at each time point remain unclear, as does potential molecular mechanisms temporal characteristics of TLE. Resting-state magnetic resonance imaging (rs-fMRI) was acquired for 84 TLE patients and 35 healthy controls (HCs). The data then used to conduct HMM analysis on rs-fMRI from an HC group order explore intricate dynamics cognitive impairment (TLE-CI). Additionally, we aim examine gene expression profiles modular using Allen Human Brain Atlas (AHBA) database. Five states were identified this study. Compared HCs, TLE-CI exhibited distinct dynamics, including fractional occupancy, lifetimes, mean dwell switch rate. Furthermore, transition probability across significantly different between (p < 0.05). reconfiguration several networks (including high-order default mode network (DMN), subcortical (SCN), cerebellum (CN). a total 1580 genes revealed be TLE, mainly enriched neuronal signaling synaptic function. This study provides new insights into characterizing neural defined by may deepen our understanding neurobiological underpinnings TLE-CI, indicating linkage configuration

Abstract The cerebellum, a subcortical structure, is traditionally linked to sensorimotor integration and coordination, although its role in cognition affective behavior, as well epilepsy, increasingly recognized. Cerebellar dysfunction patients with epilepsy can result from genetic disorders, antiseizure medications, seizures, seizure‐related trauma. Impaired cerebellar function, regardless of cause, cause ataxia (imbalance, impaired unsteady gait), tremor, gaze‐evoked nystagmus, slow gaze pursuit saccade accuracy, speech deficits (slurred, scanning, or staccato). We explore how contribute reviewing data on genetic, infectious, neuroinflammatory disorders. Evidence comes animal studies human neuropathology structural magnetic resonance imaging (MRI), functional diffusion tensor MRI, positron emission single photon computerized tomography, depth electrode electro‐encephalography studies. lesions infrequently focal motor, autonomic, bilateral tonic–clonic seizures. Electrical stimulation cerebellum has yielded conflicting results efficacy, methodological issues confound interpretation. Epilepsy‐related comorbidities including cognitive falls, sudden unexpected death may also be impacted by dysfunction. discuss drive epilepsies, seizure therapies dysfunction, our understanding epilepsy‐related through basic neuroscience, animals models patient advance improve outcomes.

Understanding the molecular mechanisms underlying generation of absence seizures is crucial for developing effective, patient-specific treatments childhood epilepsy (CAE). Currently, one-third patients remain refractive to antiseizure medications (ASMs), previously called antiepileptic drugs (AEDs), available treat CAE. Additionally, these ASMs often produce serious side effects and can even exacerbate symptoms in some patients. Determining precise cellular directly responsible causing this type has proven challenging as they appear be complex multifactorial with different genetic backgrounds. Aberrant neuronal activity CAE may caused by several that are not fully understood. Thus, dissecting causal factors could targeted development precision medicines without remains a high priority ultimate goal field research. The aim review highlight our current understanding potential causative seizure generation, based on latest research using cutting-edge technologies. This information will important identifying targets future therapeutic intervention.

The unclear propagation pathway of seizures is one the main reasons for failure surgical treatment, and process involves directional brain networks. However, few network analysis techniques have successfully traced specific seizure pathways. This study proposed a stepwise transfer entropy (STE) approach to describe effective connections in STE technique applied stereoelectroencephalography (SEEG) data collected from patients with epilepsy, which can identify characteristic regions connected seed at different link-step levels. Importantly, underlying TE-based construction method accurately obtain electrode-to-electrode connections, capture interactions between electrodes connection level. Finally, simulation clinical were used evaluate according similarity, confusion matrix, accuracy recall. We three datasets: dataset, public dataset. compared results methods multiple datasets, captured node changes epilepsy patients, effectively identified early late nodes, determined pathway. Moreover, superior other all evaluation indices, achieving 97.9% 0.93 similarity. Compared existing methods, has significant advantages tracking simple quickly performs calculations, making it an easy-to-use promising determining pathways settings.
