For more than 20 years, we have known that Ca 2+ /calmodulin-dependent protein kinase (CaMKII) activation is both necessary and sufficient for the induction of long-term potentiation (LTP). During this time, tremendous effort has been spent in attempting to understand how CaMKII gives rise phenomenon. Despite such efforts, there much be learned about molecular mechanisms involved LTP downstream activation. In review, highlight recent developments shaped our current thinking underlying discuss important questions remain field.

A current issue in the research of augmentation brain functions using transcranial electrical stimulation (tES) is diversity and inconsistency outcome results. Similar studies often report different results, depending on parameters tasks used. Such inconsistencies have led to significant doubts about efficacy method broader scientific community, despite its promising potential for patient recovery treatment. Evidence large variability individual cortical excitability response tES suggests that may affect individuals differently, subject's age, gender, state, hormonal levels, pre-existing regional excitability. Certain factors might even lead reversal polarity-dependent effects, therefore crucial implications neurorehabilitation cognitive enhancement. Most elementary behaviors such as moving the arm to grasp an object or walking into next room explore a museum evolve on time scale of seconds; in contrast, neuronal action potentials occur few milliseconds. Learning rules brain must therefore bridge gap between these two different scales. Modern theories synaptic plasticity have postulated that co-activation pre- and postsynaptic neurons sets flag at synapse, called eligibility trace, leads weight change only if additional factor is present while set. This third factor, signaling reward, punishment, surprise, novelty, could be implemented by phasic activity neuromodulators specific inputs special events.
