Decisions are often made by accumulating ambiguous evidence over time. The brain’s arousal systems activated during such decisions. In previous work in humans, we found that evoked responses of decisions reported rapid dilations the pupil and track a suppression biases accumulation decision-relevant (de Gee et al., 2017). Here, show this arousal-related decision bias acts on both conservative liberal biases, generalizes from humans to mice, perceptual memory-based challenging sound-detection tasks, impact spontaneous or experimentally induced choice was reduced under high phasic arousal. Similar occurred when drawn memory. All these behavioral effects were explained biases. Our results point general principle interplay between decision-making.

Many large-scale functional connectivity studies have emphasized the importance of communication through increased inter-region correlations during task states. Dopamine is required for working memory, but how it modulates the large-scale cortex unknown. Here, we report that dopamine receptor density per neuron, measured by autoradiography, displays a macroscopic gradient along macaque cortical hierarchy. Significance Grasping objects is something primates do effortlessly, but how does our brain coordinate such a complex task? Multiple areas across the parietal and frontal cortices of macaque monkeys are essential for shaping hand during grasping, we lack comprehensive model grasping from vision to action. In this work, show that multiarea neural networks trained reproduce arm control required using visual features also reproduced dynamics in regions relationships between areas, outperforming alternative models. Simulated lesion experiments revealed unique deficits paralleling lesions specific circuit, providing these work together drive behavior.

Correlates of sensory stimuli and motor actions are found in multiple cortical areas, but such correlates do not indicate whether these areas causally relevant to task performance. We trained mice discriminate visual contrast report their decision by steering a wheel. Widefield calcium imaging Neuropixels recordings cortex revealed stimulus-related activity (VIS) frontal (MOs) widespread movement-related across the whole dorsal cortex. Optogenetic inactivation biased choices only when targeted at VIS MOs,proportionally each site's encoding stimulus, times corresponding peak stimulus decoding. A neurometric model based on summing subtracting MOs successfully described behavioral performance predicted effect optogenetic inactivation. The brain functions through coordinated activity among distributed regions. Wide-field calcium imaging, combined with improved genetically encoded indicators, allows sufficient signal-to-noise ratio and spatiotemporal resolution to afford a unique opportunity capture cortex-wide dynamics on moment-by-moment basis in behaving animals. Recent applications of this approach have been uncovering cortical at unprecedented scales during various cognitive processes, ranging from relatively simple sensorimotor integration more complex decision-making tasks. In review, we will highlight recent scientific advances enabled by wide-field imaging mice. We then summarize several technical considerations future opportunities for uncover large-scale circuit dynamics.
