Abstract Accurate goal-directed behavior requires the sense of touch to be integrated with information about body position and ongoing motion 1,2,3 . Behaviors like chewing, swallowing speech critically depend on precise tactile events a rapidly moving tongue 4,5 , but neural circuits for dynamic touch-guided control are unknown. Using high speed videography, we examined 3D lingual kinematics as mice drank from water spout that unexpectedly changed during licking, requiring re-aiming in response subtle contact left, center or right surface tongue. Mice both re-aim ensuing licks. Surprisingly, was unaffected by photoinactivation sensory, premotor motor cortices, impaired lateral superior colliculus (latSC). Electrophysiological recordings identified latSC neurons mechanosensory receptive fields were anchored tongue-centered, head-centered conjunctive reference frames. Notably, also encoded before contact, important tongue-to-head based coordinate transformations underlying accurate aiming. Viral tracing revealed sensory inputs trigeminal nucleus, optical microstimulation topographic map aiming These findings demonstrate first time relies collicular mechanosensorimotor map, analogous visuomotor maps associated visually-guided orienting across many species.

Pleasurable touch is paramount during social behavior, including sexual encounters. However, the identity and precise role of sensory neurons that transduce remain unknown. A population labeled by developmental expression G protein-coupled receptor Mrgprb4 detects mechanical stimulation in mice. Here, we study relevance Mrgprb4-lineage reveal these are required for receptivity sufficient to induce dopamine release brain. Even isolation, optogenetic through back skin a conditioned place preference striking dorsiflexion resembling lordotic copulatory posture. In absence neurons, female mice no longer find male mounts rewarding: supplanted aggression coincident decline nucleus accumbens. Together, findings establish initiate skin-to-brain circuit encoding rewarding quality touch.

Cancer models, which are biologically representative of patient tumors, can predict the treatment responses and help determine most appropriate cancer for individual patients. Here, a point-of-care testing system called acoustically bioprinted patient-derived microtissues (PDMs) that model invasion response in patients with colorectal (CRC), is reported. The PDMs composed tumors healthy organoids be precisely arranged by acoustic bioprinting approach recapulating primary tissue's architecture. Particularly, these tumor efficiently generated apprehend histological, genomic, phenotypical characteristics tumors. Consequently, allow physiologically relevant vitro drug (5-fluorouracil) screens, thus predicting paired patient's to chemotherapy. A correlation between organoid speed normalized spreading further established. It provides quantitative indicator doctors make better decisions on ultimate anus-preserving operation extremely low CRC Thus, combing cultures, this method may open an avenue establish complex 3D tissue models precision personalized medicine.

The feedback projections from cortical layer 6 (L6CT) to the sensory thalamus have long been implicated in playing a primary role gating signaling but remain poorly understood. To causally elucidate full range of effects these projections, we targeted silicon probe recordings whisker thalamocortical circuit awake mice selectively expressing Channelrhodopsin-2 L6CT neurons. Through optogenetic manipulation neurons, multi-site electrophysiological recordings, and modeling circuitry, establish neurons as dynamic modulators ongoing spiking ventral posteromedial nucleus (VPm), either suppressing or enhancing VPm depending on neurons' firing rate synchrony. Differential across excitatory inhibitory sub-populations point an overall influence excitability that could profound implications for regulating ethologically relevant conditions.

Neocortical sensory areas have associated primary and secondary thalamic nuclei. While nuclei transmit information to cortex, remain poorly understood. We recorded juxtasomally from somatosensory (POm) visual (LP) of awake mice while tracking whisking pupil size. POm activity correlated with whisking, but not precise whisker kinematics. This coarse movement modulation persisted after facial paralysis thus was due reafference. phenomenon also continued during optogenetic silencing motor cortex lesion superior colliculus, ruling out a efference copy mechanism. Whisking dilation were strongly correlated, possibly reflecting arousal. Indeed LP, which is part the system, tracked equally well, further indicating that does encode per se. Abstract Neuronal responses during behavior are diverse, ranging from highly reliable ‘classical’ to irregular ‘non-classically responsive’ firing. While a continuum of response properties is observed across neural systems, little known about the synaptic origins and contributions diverse network function, perception, behavior. To capture heterogeneous measured auditory cortex rodents performing frequency recognition task, we use novel task-performing spiking recurrent incorporating spike-timing-dependent plasticity. Reliable units contribute differentially task performance via output connections, respectively. Excitatory plasticity shifts distribution while inhibition constrains its diversity. Together both improve with full engagement. The same local patterns inputs predict cortical neurons in vivo whole-cell recordings Thus, function emerge rules.

Abstract Primary sensory cortex has long been believed to play a straightforward role in the initial processing of information. Yet, superficial layers overall are sparsely active, even during stimulation; additionally, cortical activity is influenced by other modalities, task context, reward, and behavioral state. Our study demonstrates that reinforcement learning dramatically alters representations among longitudinally imaged neurons mouse primary somatosensory cortex. Learning an object detection recruits previously unresponsive neurons, enlarging neuronal population sensitive touch choice. Cortical responses decrease upon repeated stimulus presentation outside task. Moreover, training improves encoding passage time, unexpected deviations trial timing elicit stronger than touches do. In conclusion, exhibit high degree learning-dependent plasticity strongly modulated non-sensory but behaviorally-relevant features, such as surprise.
