The claustrum (CLA), a subcortical nucleus in mammals, essentially composed of excitatory projection neurons and known for its extensive connections with the neocortex, has recently been associated variety functions ranging from consciousness to impulse control. However, research on CLA challenging due difficulties specifically comprehensively targeting neuronal populations. In various cases, this limitation led inconsistent findings lack reliable data. present work, we describe expression profile Smim32 gene, which is almost exclusively transcribed endopiriform nucleus, as well inhibitory thalamic reticular nucleus. Leveraging unique pattern, developed series Cre- Flippase-expressing knockin BAC transgenic mouse lines different profiles. With these novel tools hand, propose new standards interrogation function.

The hypothalamic neuropeptide, oxytocin (Oxt), has been the focus of research for decades due to its effects on body physiology, neural circuits, and various behaviors. Oxt elicits a multitude actions mainly through receptor, receptor (OxtR). Despite past understand central projections neurons OxtR- coupled signaling pathways in different brain areas, it remains unclear how this nonapeptide exhibits such pleiotropic while integrating external internal information. Most reviews field either neuroanatomy Oxt-OxtR system, or functional specific areas. Here, we provide review by wide connectivity their downstream circuits with OxtR expression mice. We categorize connected regions into three modules that regulate state, somatic visceral, cognitive response. Each module contains process distinct behavioral effects. Broad innervations (e.g., basal ganglia motor behavior) enable exert coordinated modulation functionally inter-connected circuits. Moreover, acts as neuromodulator neuromodulations broadly control overall state brain. Lastly, discuss mismatch between across mouse In summary, brings forth circuit-based analysis whole light release provides perspective guide future studies.

The claustrum, a structure having extensive connectivity with the rest of brain and being involved in many high-cognitive processes, is still one least understood parts mammalian nervous system. Accelerated advancement genetic tools for rodents last decade have resulted breakthroughs about its interaction cortical subcortical regions, while human/primate studies been invaluable revealing effects on conscious behaviour. The claustrum is thought to be one of the most highly interconnected forebrain structures but its organizing principles have yet fully explored at level single neurons. Here, we investigated identity, connectivity, and activity identified neurons understand how structure’s unique convergence input divergence output support binding information streams. We found that in communicate with each other across efferent projection-defined modules which were differentially innervated by sensory frontal cortical areas. Individual responsive inputs from more than region a cell-type projection-specific manner, particularly between areas cortex. In vivo imaging axons revealed responses both unimodal multimodal stimuli. Finally, chronic silencing specifically reduced animals’ sensitivity These findings view fundamentally integrative structure, consolidating around cortex redistributing it following local computations.
