The incidence of inflammatory bowel disease (IBD) and the associated risk colon cancer are increasing globally. Traditional Chinese medicine (TCM) treatment has unique advantages. Sishen Pill, a common patented drug used to treat abdominal pain diarrhea, consists mainly Psoraleae Fructus, Myristicae Semen, Euodiae Schisandra Chinensis. Modern research confirmed that Pill its active secondary metabolites, such as psoralen, myristicin, evodiamine, schisandrin, can improve intestinal inflammation exert antitumor pharmacological effects. Common mechanisms in treating IBD include regulating inflammation-related signaling pathways nuclear factor-kappa B, mitogen-activated protein kinase, phosphatidylinositol 3-kinase, NOD-like receptor heat domain-related 3, wingless-type MMTV integration site family; NF-E2-related factor 2 hypoxia-inducible 1α inhibit oxidative stress; mitochondrial autophagy endoplasmic reticulum immune cell differentiation function through Janus kinase/signal transducer activator transcription pathway; improving gut microbiota barrier. Overall, existing evidence suggests potential pill suppress inflammation-to-cancer transformation. However, large-scale randomized controlled clinical studies on safety these applications urgently required.

Abstract Background Colorectal cancer (CRC) remains a significant cause of morbidity and mortality worldwide. In patients with inflammatory bowel disease, who have twice the risk developing CRC, chronic inflammation has been recognized to contribute colitis-associated (CAC) development. Jacalin, lectin extracted from jackfruit seeds, shown recognize altered glycosylation exert antiproliferative cytotoxic effects in CRC. However, its activity CAC unknown. Herein, we sought investigate jacalin progression using dextran sulfate sodium (DSS) azoxymethane (AOM) mouse model. Methods Colitis-associated induction was performed male C57BL/6 mice by an intraperitoneal injection AOM, followed 3 cycles 2.5% DSS diluted drinking water for 7 days, intercalated 2 weeks normal water. After 1 week daily pretreatment, were orally treated phosphate-buffered saline (control group), 100 or 500 µg three times additional 11 weeks. Results We showed that jacalin-treated presented tumors reduced volumes mean size compared control group. addition, both doses number proliferating cells (Ki-67 positive cells) tumor tissues, while higher dose (500 µg) also similar effect “normal-appearing” colonic crypts. Jacalin treatment attenuated clinical scores inflammations, which accompanied reduction intestinal and/or tumoral production IL-1β, IL-23, IL-17. Conclusions Collectively, our findings demonstrated suppresses development, highlighting anti-inflammatory antitumoral role AOM/DSS-induced

Colorectal cancer is one of the most common malignant tumors, with numerous studies highlighting role inflammation in its onset and progression. Cytokines such as IL-6 TNF-a play a crucial sustaining inflammation, contributing to transformation cells. The dietary inflammatory index, which reflects intake pro-inflammatory nutrients, associated an increased risk developing colorectal cancer. Serum cytokine concentrations may serve biomarkers for assessment, while modifications aimed at reducing can significantly impact both prevention therapy. This knowledge opens possibilities personalized approach treatment

Over the past 24 years, nuclear receptor-binding SET domain protein 3 (NSD3) has emerged as a critical regulator of cellular physiological processes. As histone methyltransferase targeting H3K36, NSD3 catalyzes addition methyl groups to residues, process that profoundly influences genome imprinting, gene transcription, and stability, thereby modulating expression. Amplification, mutations, fusion events involving have been strongly linked pathogenesis various cancers, highlighting its role key tumorigenesis. This review provides an overview general structure biological functions NSD3, followed by analysis NSD3's in relation hallmarks cancer described Hanahan Weinberg. Targeting become major focus research, with significant efforts directed toward development clinical application inhibitors. However, challenges related specificity selectivity hindered progress this area. Despite these obstacles, successful advancement other inhibitors provided encouragement researchers, driving active pursuit NSD3-targeted therapies for treatment.

Hesperetin, a flavonoid predominantly present in citrus fruits, exhibits significant intervention effects on both the initiation and progression of gastric cancer. However, specific mechanisms underlying this effect remain unclear. To investigate interventional role hesperetin N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced exosomes inducing carcinogenesis. Bioinformatics technology was used to identify critical molecular components hesperetin-mediated inhibition MNNG induced carcinogenesis through exosomal circular RNA. Colo-rectal cancer (CRC) is undoubtedly one of the most severe complications inflammatory bowel diseases (IBD). While sporadic CRC develops from a typical adenoma-carcinoma sequence, IBD-related follows different and less understood pathways its pathophysiological mechanisms were not completely elucidated. In contrast to chronic inflammation, which nowadays well-recognised drive towards neoplastic transformation in IBD, only recently was gut microbiota demonstrated interfere with both inflammation processes immune-mediated anticancer surveillance. Moreover, role appears particularly complex intriguing when also considering multifaceted interactions multiple environmental stimuli, notably pathologies such as diabetes obesity, lifestyle (diet, smoking) vitamin intake. this review, we presented comprehensive overview on current evidence influence CRC, particular mutual interconnections environment.
