Abstract Objectives Rheumatoid arthritis (RA) is an inflammatory and autoimmune disease. Studies over the past two decades suggest that statins have a beneficial impact on complications associated with RA. These include RA disease activity risk for cardiovascular diseases (CVD). This review aims to discuss efficacy of statin therapy in Key findings The current evidence suggests statins' immunomodulatory antioxidant properties significantly reduce response patients In patients, CVD reduced by treatment, discontinuation increased risk. Summary combined effect improving vascular function, lowering lipid levels, reducing inflammation responsible decreased all-cause mortality users. Myocardial ischemia-reperfusion injury (MI/RI) is a critical challenge for acute myocardial infarction therapy, as there currently no ideal drug available. Glycogen synthase kinase 3 beta (GSK-3β) serves an promising therapeutic target treating MI/RI. Our previous studies have demonstrated that Ilex pubescens ameliorates The purpose of this study to evaluate the efficacy and potential mechanism screened GSK-3β inhibitor from against Three-dimensional-quantitative structure-activity relationship (3D-QSAR) modeling, molecular docking, oxygen glucose deprivation/reperfusion (OGD/R) left anterior descending (LAD) artery ligation-induced MI/RI mice model, western blotting analysis were used screen investigate protective mechanism. Here, we Pubescenoside D (PBD) with IC50 value 0.3769 μM pubescens, using 3D-QSAR assay verification. Ile217, Leu88, Phe93, Phe67 are key binding sites PBD GSK-3β. protects cardiomyocytes in vitro vivo. Further show inhibits mitochondrial permeability transition pore (mPTP) opening by preventing GSK-3β-mediated dissociation hexokinase2 (HK2) outer membrane mitochondria enhances mitophagy suppressing activity, subsequently reducing cardiomyocyte apoptosis. findings shed light on agent treatment targeting

Abstract Protein tyrosine phosphatase mitochondrial 1 (PTPMT1), is a member of the protein superfamily localized on inner membrane, and regulates biosynthesis cardiolipin. Given important position PTPMT1 in function metabolism, pharmacological targeting considered promising manner disease treatments. In this study, we mainly investigated role hepatocellular carcinoma (HCC) ferroptosis, new type cell death accompanied by significant iron accumulation lipid peroxidation. Herein, inhibition was induced alexidine dihydrochloride (AD, dibiguanide compound). Human HCC lines with knockout overexpression were established using CRISPR/Cas9 lentiviral transduction methods, respectively. The regulating ferroptosis evaluated vitro vivo. Our results indicated that PTPMT1, facilitated AD treatment, heightens susceptibility to cystine deprivation-ferroptosis, treatment promoted conversion from ferritin-bound Fe 3+ free 2+ , which contributed labile pool cytoplasm. Meanwhile, also formation both swollen mitochondria donut mitochondria, enhanced metabolism process form succinate fumarate tricarboxylic acid (TCA) cycle, increased sensitivity cells deprivation-induced ferroptosis. total, our work reveals close association cysteine providing novel insight into chemotherapy strategies against human HCC.

Abdominal aortic aneurysm (AAA) is a prevalent condition which causes progressive growth and rupture of wall with high death rate. Several studies have found that treatment statins may decrease the progress AAA risk by suppressing inflammatory mediators, decreasing oxidative stress, inhibiting mechanisms involved in extracellular matrix (ECM) degradation. Moreover, some reported prehospital therapy can mortality after surgery. The novelty this paper different including those performed humans animals were reviewed potential an effect on summarized. Overall, evidence suggested association between improvement AAA.

The cardiovascular benefit of statins is well established. However, only 20% high-risk patients remain adequately adherent after 5 years treatment. Among reasons for discontinuation, statin associated-muscle pain symptoms are the most prevalent. Aim present study was to evaluate impact high dose atorvastatin on skeletal muscle mitochondrial activity, aerobic and anaerobic exercise, axonal excitability in a murine model atherosclerosis. ApoE-/- mice were fed 12 weeks high-fat high-cholesterol diet alone or containing (40 mg/Kg/day). Outcomes evaluation functionality, locomotion, grip test, (compound action potential recording analysis Aα motor propioceptive, Aβ mechanoceptive C nociceptive fibres). Atorvastatin led reduction biogenesis ATP production. It did not affect muscular strength but time-dependent impairment. altered responsiveness fibres, respectively, fibres. These findings point out mild sensitization mechanical, tactile sensitivity. In conclusion, although prevalence side effects from may be overestimated, understanding underlying mechanisms can help improve therapeutic approach reassure adherence needing-to-be-treated.
