Abstract The α4β2 nicotinic acetylcholine receptor (nAChR), an ionophore, has been suggested to signal through metabotropic pathways and interact with other families, such as dopamine receptors. In this study, the interaction between nAChR receptors was investigated in vivo vitro studies. Nicotine exposure adolescent rats is known induce a sustained increase nicotine's rewarding effects which assessed by conditioned place preference (CPP) assay. expression levels of D 2 /D 3 (D R, R) increased after nicotine treatment. To determine these two treatment, newly developed ligand high selectivity for R used radioligand binding Although both enhanced during adolescence, only elevated level persisted into adulthood. experiments conducted on mice, knockout mice showed significantly lower CPP scores adulthood compared wild‐type mice. Cellular studies that attributed promoter activity, regulated signaling cascade composed Src, Syk, PKC, NF‐κB. These results demonstrate pathway involved also suggest how reward initiated adolescence could relapse long abstinence period. Given significance addiction, study thought offer novel mechanistic perspective understanding relapse. image

Parkinson’s disease (PD) is a progressive neurodegenerative characterized by resting tremor, bradykinesia, rigidity, and postural instability that also includes non-motor symptoms such as mood dysregulation. Dopamine (DA) the primary neurotransmitter involved in this disease, but cholinergic imbalance has been implicated. Current intervention PD focused on replenishing central DA, which provides remarkable temporary symptomatic relief does not address neuronal loss progression of disease. It well established nicotinic receptors (nAChRs) can regulate DA release nicotine itself may have neuroprotective effects. Recent studies identified nAChRs nonneuronal cell types, including glial cells, where they inflammatory responses. Given crucial role neuroinflammation dopaminergic degeneration involvement microglia astrocytes response, provide novel therapeutic target prevention and/or treatment PD. In review, following brief discussion PD, we focus cells and, specifically, their pathology treatment.

Glutamate is the major excitatory neurotransmitter in central nervous system. Glutamatergic transmission can be mediated by ionotropic glutamate receptors (iGluRs), which mediate rapid synaptic depolarization that associated with Ca2+ entry and activity-dependent change strength of transmission, as well metabotropic (mGluRs), slower postsynaptic responses through recruitment second messenger systems. A wealth evidence reported over last three decades has shown this dogmatic subdivision between iGluRs mGluRs may not reflect actual physiological signaling mode iGluRs, i.e., α-amino-3-hydroxy-5-methyl-4-isoxasolepropionic acid (AMPA) (AMPAR), kainate (KARs), N-methyl-D-aspartate (NMDA) (NMDARs). Herein, we review available supporting notion canonical recruit flux-independent pathways only neurons, but also brain astrocytes cerebrovascular endothelial cells. Understanding versatility exert a profound impact on our understanding glutamatergic synapses. Furthermore, it shed light novel neuroprotective strategies against disorders.

Nicotinic acetylcholine receptors (nAChRs) are widely expressed in a variety of cell types and involved multiple physiological regulatory mechanisms cells, tissues systems. Increasing evidence suggests that the α5 nicotinic receptor (α5-nAChR), encoded by CHRNA5 gene, is one key mediator lung cancer development immune responses. Several studies have shown it regulator stimulates processes via various signaling pathways, including STAT3 cancer. In addition, α5-nAChR has profound effect on response through immune-related factor pathways. this review, we focus perspectives progression, which indicates targeting could provide novel anticancer therapy strategies for

Nicotinic acetylcholine receptors (nAChRs) are a family of ligand-gated ion channel that contribute to cognition, memory, and motor control in many organisms. The pharmacological targeting these receptors, using small molecules or peptides, presents an important strategy for the development drugs can treat human diseases, including neurodegenerative disorders. Aplysia californica binding protein (Ac-AChBP) is structural surrogate nAChR with high homology extracellular ligand domain homopentameric nAChRs. In this study, we optimized protein-painting-based mass spectrometry identify regions interaction between Ac-AChBP several ligands. Using molecular dyes adhere surface solubilized complex, identified amino acid residues constitute contact site within α-bungarotoxin, choline, nicotine, amyloid-β 1–42. By integrating innovation painting computational modeling, present new experimental tool analyzing interactions nAChR.
