Abstract: Pyroptosis is a novel pro-inflammatory mode of programmed cell death that differs from ferroptosis, necrosis, and apoptosis in terms its onset regulatory mechanisms. dependent on cysteine aspartate protein hydrolase (caspase)-mediated activation GSDMD, NLRP3, the release cytokines, interleukin-1 (IL-1β), interleukin-18 (IL-18), ultimately leading to death. Non-coding RNA (ncRNA) type does not encode proteins gene transcription but plays an important role other post-transcriptional links. NcRNA mediates pyroptosis by regulating various related factors, which we termed signaling pathway. Previous researches have manifested closely development liver diseases, essential for injury, alcoholic fatty disease (ALD), non-alcoholic (NAFLD), fibrosis, cancer. In this review, attempt address ncRNA-mediated pathway above diseases their pathogenesis recent years, briefly outline TCM (Traditional Chinese Medicine) intervene modulating pyroptosis, will provide strategy find new therapeutic targets prevention treatment future. Abstract Liver fibrosis is a chronic liver lesion caused by excessive deposition of the extracellular matrix after damage, resulting in fibrous scarring tissue. The progression partially influenced gut microbiota. Chitosan can play therapeutic role regulating microbiota based on ‘gut–liver axis’ theory. Salvianolic acid B inhibit development inhibiting activation hepatic stellate cells and reducing production matrix. In this study, effect chitosan combination with salvianolic was investigated mouse model. results showed that better than drug alone, improving AST/ALT levels expression α‐SAM, COL I, IL‐6 other related genes. It improved structure increased abundance beneficial bacteria such as Lactobacillus. above could provide new ideas for clinical treatment fibrosis.

Distant metastasis is a major cause of treatment failure in cancer patients and key challenge to improving care today. We hypothesized that enhancing anti-cancer immune response inhibiting circulating tumor cells (CTCs) adhesion transendothelial migration through synergistic multi-target approaches may effectively prevent metastasis. “Fuyuan Decoction” (FYD) traditional Chinese medicine compound widely used postoperative patients, but its underlying mechanism remains unclear. In this work, we systematically elucidated the molecular by which FYD prevents multi-compound synergies vitro vivo . significantly prevented at non-cytotoxic concentrations suppressing CTCs endothelial their subsequent migration, as well response. Mechanistically, interrupts vascular endothelium TNF-α-induced CAMs expression via regulation NF-κB signaling pathway cells. inhibits invasion EMT, PI3K/AKT FAK pathways. Moreover, enhances increasing population Tc NK peripheral system. addition, chemical composition was determined UPLC-HRMS, results indicated multiple compounds treatment. This study provides modern medical basis for application prevention metastasis, suggesting multi-drug therapy be one most effective ways

Liver fibrosis is a complex process driven by various factors and key feature of chronic liver diseases. Its essence tissue remodeling caused excessive accumulation collagen other extracellular matrix. Activation hepatic stellate cells (HSCs), which are responsible for production, plays crucial role in promoting the progression fibrosis. Abnormal expression signaling pathways, such as TGF-β/Smads pathway, contributes to HSCs activation. Recent studies have shed light on these providing valuable insights into development Here, we will review six pathways that been studied more recent years.

Liver fibrosis is a common complication of chronic liver disease, significantly affecting patients’ quality life and potentially leading to cirrhosis hepatocellular carcinoma. Despite advancements in modern medicine, the treatment remains limited challenging. Thus, identifying new therapeutic strategies great clinical importance. Signaling pathways related play crucial regulatory role immune response inflammation. Aberrant activation specific pathways, such as NF-[Formula: see text]B signaling pathway, results overexpression genes associated with inflammation fibrosis, thereby promoting progression fibrosis. Chinese medicine offers unique potential advantages approach. Recent studies have increasingly demonstrated that certain medicines can effectively treat by regulating relevant pathways. The active ingredients these inhibit hepatic inflammatory responses fibrotic processes interfering thus reducing severity This paper aims investigate mechanisms treating its modulation Additionally, it discusses prospects application treatments provides valuable references for further research practice.
