Chronic inflammation is a common foundation for the development of many non-communicable diseases, particularly diabetes, atherosclerosis, and tumors. The activation axis involving Advanced Glycation End products (AGEs) their receptor RAGE key promotive factor in chronic process, influencing pathological progression these diseases. accumulation AGEs body results from an increase glycation reactions oxidative stress, especially pronounced individuals with diabetes. By binding to RAGE, activate signaling pathways such as NF-κB, promoting release inflammatory factors, exacerbating cell damage inflammation, further advancing formation atherosclerotic plaques tumor development. This review will delve into molecular mechanisms by which AGEs-RAGE activates aforementioned well strategies inhibit axis, aiming slow or halt related includes inhibitors, antagonists, interventions targeting upstream downstream pathways. Additionally, early detection levels expression biomarkers provides new avenues prevention treatment

Abstract Enhancement of malignant cell immunogenicity to relieve immunosuppression lung cancer microenvironment is essential in treatment. In previous study, we have demonstrated that dihydroartemisinin (DHA), a kind phytopharmaceutical, effective inhibiting cells and boosting their immunogenicity, while the initial target DHA's intracellular action poorly understood. The present in‐depth analysis aims reveal influence DHA on highly expressed TOM70 mitochondrial membrane cancer. affinity was analyzed by microscale thermophoresis (MST), pronase stability, thermal stability. functions underlying mechanism were investigated using western blots, qRT‐PCR, flow cytometry, rescue experiments. inhibition resulted mtDNA damage translocation cytoplasm from mitochondria due disruption homeostasis. Further ex vivo findings also showed cGAS/STING/NLRP3 signaling pathway activated thereby underwent pyroptosis, leading enhanced presence DHA. Nevertheless, DHA‐induced mobilization synchronously attenuated when replenished. Finally, possess potent anti‐lung efficacy vitro vivo. Taken together, these data confirm an important for disturb homeostasis, which further activates STING arouses pyroptosis strengthen against thereupon. study provides vital clues phytomedicine‐mediated anti‐tumor therapy.

The consumption of wampee has traditionally been utilized to alleviate gastrointestinal inflammation and associated disorders; however, its exact mechanism remained unknown. aim this study was elucidate the therapeutic efficacy underlying polyphenol extract (WPE) in dextran sulfate sodium (DSS)-induced ulcerative colitis (UC). findings revealed that WPE alleviated diverse symptoms UC, regulated various inflammatory cytokines, effectively protected colon tissue structure barrier integrity, thereby inhibiting LPS translocation. Moreover, restored richness diversity gut microbiota optimized at phylum genus levels, causing a notable improvement short- chain fatty acid (SCFA) metabolism, particularly acetic acid, propionic butyric acid. Consequently, demonstrated suppress LPS-induced TLR4-p38 MAPK/NF-κB signaling pathway by modulating SCFA metabolism. These provided theoretical basis for use as potential functional natural food UC.

Abstract Objectives This investigation aimed to evaluate the efficacy, key active ingredients, and mechanisms of Zuoqing San (ZQS), a traditional Chinese medicine formula, in managing sigmoid ulcerative colitis (SUC). Methods A cohort 126 participants was recruited treated with ZQS at daily dosage 100 mL for 12 weeks. The primary endpoint percentage subjects achieving favorable treatment outcomes. Liquid chromatography-mass spectrometry employed identify principal ingredients ZQS. Network pharmacology utilized predict central targets its ingredients. Protein-protein interaction networks, Gene Ontology enrichment analyses, Kyoto Encyclopedia Genes Genomes pathway analyses were conducted identified core targets. Finally, molecular dynamics simulations performed. Results Following weeks therapy, withdrawal rate 7.93 %, outcomes observed 31.03 % 4 weeks, 66.37 8 (Odds Ratio: 1.54, 95 Confidence Interval: 0.41–1.83), 68.10 1.86, CI: 0.32–1.27). comprises 31 chemical constituents. Key within protein-protein network included TNF, AKT1, IL6, IL1β, PTGS2, TP53, JUN, MMP9, CASP3, HIF1A, BCL1. Pathway analysis indicated primarily impacts NF-kB, IL-17 signaling pathways. Molecular simulation results showed oxymatrine, cynarin had higher affinity TNF respectively. Conclusions research elucidates components potential multicomponent–multitarget–multipathway pharmacological mechanisms, demonstrating promising therapeutic SUC management.
