ABSTRACT Combination therapy offers a promising advantage because they target multiple pathways involved in the pathogenesis and progression of Alzheimer's disease. This study aimed to investigate synergistic effect donepezil each vitamin B12, B6 B1 on aluminium chloride (AlCl 3 )‐induced neurotoxicity rats. Fifty‐four rats were divided into nine groups six. Group I received distilled water, group II—AlCl (100 mg/kg), III‐ AlCl mg/kg) + Donepezil (10 IV‐AlCl Vitamin B12 (5 V‐AlCl VI‐AlCl VII‐AlCl VIII‐AlCl IX‐AlCl mg/kg). Treatment lasted for 40 days. Biochemical analyses brain revealed that chronic administration significantly increased level inflammatory cytokines, caspase 9, malondialdehyde, nitric oxide acetylcholinesterase (AChE) activity, while causing significant decrease superoxide dismutase (SOD) Nrf2, reduced glutathione, dopamine norepinephrine. Results also showed caused cognitive impairment as indicated by reduction percentage alternation index hypo‐activity towards novel object animals treated with only. Moreover, results from biochemical evaluations treatment donepezil, B vitamins combination attenuated deficits dopamine, norepinephrine, SOD reducing elevated levels oxide, AChE, 9 markers. These observations corroborated result histopathological evaluation. The proved be more effective than monotherapy. conferred protection against –induced neurotoxicity, however, performed better neuroprotective indices was amongst three therapies this study.

Aging, an inevitable physiological process leading to a progressive decline in bodily functions, has been abundantly researched domain with studies attempting slow it down and reduce its debilitating effects. Investigations into the cellular molecular pathways associated aging have allowed formulation of therapeutic strategies. Of these, caloric restriction (CR) implicated for role promoting healthy by modulating key targets like Insulin/IGF-1, mTOR, sirtuins. However, CR requires dedication commitment strict regimen which poses difficulty maintaining consistency. To maneuver around cumbersome diets, Caloric Restriction Mimetics (CRMs) emerged as promising alternatives mimicking beneficial effects CR. This review elucidates foundations enabling CRMs rapamycin, metformin, resveratrol, spermidine, many more function suitable anti-aging molecules. Moreover, explores clinical trials (retrieved from clinicaltrials.gov database) aimed at demonstrating efficacy effective candidates against age-related neurodegenerative diseases such Alzheimer's disease Parkinson's disease.

Jambolan fruit extract and choline were investigated for Aluminum tri chloride (AlCl3)-induced Alzheimer's disease in rats. Thirty-six male "Sprague Dawley" rats weighing (150 ± 10 g) allocated into six groups; the first group was fed a baseline diet served as negative control. (AD) induced Group 2 by oral administration of AlCl3 (17 mg/kg body weight) dissolved distilled water (served positive control). Rats 3 orally supplemented concomitantly with both 500 BW an ethanolic jambolan once daily 28 days weight). 4: Rivastigmine (RIVA) aqueous infusion (0.3 BW/day) given to reference drug supplementation days. 5 treated (1.1 g/kg) 6 1.1 g/kg test additive effects concurrently bw) Body weight gain, feed intake, efficiency ratio, relative brain, liver, kidney, spleen calculated after trial. Brain tissue assessment analyzed antioxidant/oxidant markers, biochemical analysis blood serum, phenolic compound fruits extracted high-performance liquid chromatography (HPLC), histopathology brain. The results showed that improved brain functions, histopathology, antioxidant enzyme activity compared group. In conclusion, administering can lower toxic impacts aluminum on

Gallic acid (GA) is a polyphenolic substance which can be sourced naturally from plant sources and also synthesized industrially. In-depth study on GA in recent years has revealed that it powerful anti-inflammatory antioxidant properties. In many disease models, been found to ameliorate oxidative stress, inflammation resultant damage. The neuroprotective properties of against numerous animal models neurodegenerative disorders have outlined by number vivo investigations. However, this present review dedicated towards providing comprehensive information the bioavailability, effects underlying mechanism activity for management disorders. demonstratively was highlighted as backbone its effect. It safe conclude antioxidant, anti-inflammatory, metal chelating proved beneficial diseases, hence use should recommended single therapy or combination with currently approved drugs these diseases.

Colorectal cancer (CRC) is the third most common with a high mortality rate worldwide. Although gallic acid and hesperidin exert anticancer activity, synergistic effects of against CRC remain elusive. This study aims to investigate therapeutic mechanism novel combination cell growth, including viability, cell-cycle-associated proteins, spheroid formation, stemness.Gallic derived from Hakka pomelo tea (HPT) were detected by colorimetric methods high-performance liquid chromatography using ethyl acetate as an extraction medium. lines (HT-29 HCT-116) treated combined extract investigated in our for viability (trypan blue or soft agar colony formation assay), cycle (propidium iodide staining), proteins (immunoblotting), stem markers (immunohistochemistry staining).Compared other methods, HPT medium exerts potent effect on inhibiting HT-29 growth dose-dependent manner. Furthermore, treatment had higher inhibitory than alone. The underlying was involved G1-phase arrest Cip1/p21 upregulation that could attenuate HCT-116 proliferation (Ki-67), stemness (CD-133), 3D assay mimicking vivo tumorigenesis.Gallic spheroids, may serve potential chemopreventive agent. Abstract Di-(2-ethylhexyl) phthalate (DEHP) is the most abundant threatening public health-induced neurotoxicity. This neurotoxicity associated with behavioral and biochemical deficits in male rats. Our study investigated neuroprotective effect of ferulic acid (FA) on rats exposed to DEHP. Thirty-two Wistar were assigned four groups. Group I control received corn oil, group II intoxicated 300 mg/kg DEHP, III DEHP + 50 FA, IV all agents administrated daily per os for 30 days. Anxiety-like behavior, spatial working memory, recognition memory assessed. Also, brain oxidative stress biomarkers, including malondialdehyde (MDA), reduced glutathione (GSH), nitric oxide (NO), superoxide dismutase (SOD), brain-derived neurotrophic factor (BDNF) as well heme oxygenase-1 (HO-1) measured. Moreover, histopathology examinations immunohistochemistry determination caspase-3 also evaluated. Furthermore, docking simulation was adapted understand inhibitory role FA NO synthase. Compared DEHP-intoxicated rats, FA-treated displayed improved cognitive a anxious state. redox state maintained increased BNDF levels. These changes confirmed by restoring normal architecture tissue decrement caspase-3. In conclusion, has potent antioxidant antiapoptotic properties that confirm activity possible prospect its therapeutic capabilities nutritional supplement value.
