Exposure to any number of stressors during the first 1000 days from conception age 2 years is important in shaping an individual’s life trajectory health and disease. Despite expanding range as well later-life phenotypes outcomes, underlying molecular mechanisms remain unclear. Our previous data strongly suggests that early-life exposure a stressor reduces capacity immune system generate subsequent generations naïve cells, while others have shown that, early stress impairs neuronal stem cells proliferate they age. This leads us “stem cell hypothesis” whereby adversity sensitive period acts through common mechanism all types by programming tissue resident progenitor cells. Furthermore, we review mechanistic differences observed fully differentiated suggest (ELA) may alter mitochondria consequently destiny these producing lifelong “supply” functionally altered

Although dysregulated stress biology is becoming increasingly recognized as a key driver of lifelong disparities in chronic disease, we presently have no validated biomarkers toxic physiology; biological, behavioral, or cognitive treatments specifically focused on normalizing processes; and agreed-upon guidelines for treating the clinic evaluating efficacy interventions that seek to reduce improve human functioning. We address these critical issues by (a) systematically describing systems mechanisms are stress; (b) summarizing indicators, biomarkers, instruments assessing response systems; (c) highlighting therapeutic approaches can be used normalize stress-related biopsychosocial also present novel multidisciplinary Stress Phenotyping Framework bring researchers clinicians one step closer realizing goal using precision medicine-based prevent treat stress-associated health problems.

Abstract Early life adversity (ELA) has been associated with an increased risk for diseases in which the immune system plays a critical role. The ELA phenotype is characterized by inflammation, impaired cellular immunity, and immunosenescence. However, data on cell-specific effects are largely absent. Additionally, stress systems health behaviors altered ELA, may contribute to generation of phenotype. present investigation tested differences relationship phenotype, parameters, individuals (n = 42) those without history (control, n 73). Relative number activation status (CD25, CD69, HLA-DR, CD11a, CD11b) monocytes, NK cells, B T their main subsets were assessed flow cytometry. was significantly reduced numbers CD69+CD8+ cells (p 0.022), HLA-DR+ CD4 CD8 < 0.001), as well CD25+CD8+ 0.036). also showed trend toward higher CCR4+CXCR3−CCR6+ cells. Taken together, our suggest elevated state particularly affected. Although several aspects related markers, neither nor health-risk explained observed group differences. Thus, does not seem be secondary alterations or behaviors, but rather primary effect early programming
