Abstract Autism spectrum disorder (ASD) is a heterogeneous, lifelong neurodevelopmental disorder. One in five infant siblings of children with ASD later receive an diagnosis. With the aim early detection and intervention, Infant Brain Imaging Study Network conducted magnetic resonance imaging (MRI) studies siblings, typically during sleep, recently cited aberrant visual brain development as potential MRI marker ASD. Event-related functional findings showed that rapid eye movements (REMs) sleep index processing endogenous information, i.e., hierarchical generation percepts. REM-locked peak activation sites neural overlap extensively those reported to be atypical autism. Autism Spectrum Disorder (ASD) is increasing, but its complete etiology still lacking. Recently, application of ketogenic diet (KD) has shown to reduce abnormal behaviors while improving psychological/sociological status in neurodegenerative diseases. However, KD role on ASD and underlying mechanism remains unknown. In this work, administered BTBR T+ Itpr3tf/J (BTBR) C57BL/6J (C57) mice reduced social deficits (p = 0.002), repetitive < 0.001) memory impairments BTBR. Oxytocin (OXT) a neuropeptide synthesized in the hypothalamic nuclei has variety of function including socio-emotional processes mammals. While neural circuits and signaling pathways central OXT converge paraventricular nucleus hypothalamus (PVN), we illuminate specific discrete PVN circuits, which connect to medial amygdala (MeA) bed stria terminalis (BnST) mouse models. The OXTPVN→BnST projections are innervated from entire portions PVN, while those OXTPVN→MeA asymmetrically posterior portion PVN. Compared with neurons B6 wild type mice, BTBR mice that recognized as behavior-based autism model exhibited defect projection. We demonstrate chemogenetic activation circuit enhances anxiety-like behavior facilitates social approach behavior, suppresses along inhibiting approach. This manipulation on proves ineffective mice. Abstract Genetic variation can alter brain structure and, consequently, function. Comparative statistical analysis of mouse brains across genetic backgrounds requires spatial, single-cell, atlas-scale data, in replicates—a challenge for current technologies.
