The global prevalence of breast cancer necessitates the development innovative prognostic markers and therapeutic strategies. This study investigated implications anoikis-related long non-coding RNAs (ARLs) in invasive (IBC), which is an area that has not been extensively explored. By integrating RNA sequence transcriptome clinical data from Cancer Genome Atlas (TCGA) database employing advanced regression analyses, we devised a novel model based on ARL scores. scores correlated with diverse clinicopathological parameters, cellular pathways, distinct mutation patterns, immune responses, thereby affecting both cell infiltration anticipated responses to chemotherapy immunotherapy. Additionally, overexpression specific lncRNA,

Colorectal cancer (CRC) is a common and deadly disease of the digestive system, but its targeted therapy hampered by lack reliable specific biomarkers. Hence, discovering new therapeutic targets agents for CRC an urgent challenging task. Here we report that carnitine palmitoyltransferase 1A (CPT1A), mitochondrial enzyme catalyzes fatty acid oxidation (FAO), potential target treatment. We show CPT1A overexpressed in cells inhibition secolignan-type compound, 2,6-dihydroxypeperomin B (DHP-B), isolated from plant Peperomia dindygulensis, suppresses tumor cell growth induces apoptosis. demonstrate DHP-B covalently binds to Cys96 CPT1A, blocks FAO, disrupts CPT1A-VDAC1 interaction, leading increased permeability reduced oxygen consumption energy metabolism cells. also reveal expression correlates with survival tumor-bearing animals exhibits anti-CRC activity vitro vivo. Our study uncovers molecular mechanism as novel inhibitor provides rationale preclinical development well strategy therapy.

The extracellular matrix (ECM) governs a wide spectrum of cellular fate processes, with particular emphasis on anoikis, an integrin-dependent form cell death. Currently, anoikis is defined as intrinsic apoptosis. In contrast to traditional apoptosis and necroptosis, integrin correlates ECM signaling intracellular cascades, describing the full process anoikis. However, frequently overlooked in physiological pathological processes well vitro research models. this review, we summarized role spanning embryonic development, organ tissue repair, inflammatory responses, cardiovascular diseases, tumor metastasis, so on. Similarly, realm stem focused functional evolution cells, offers potential solution various challenges, including culture models, therapy, transplantation, engineering applications, which are largely based regulation by More importantly, regulatory mechanisms molecular will provide new strategies for therapeutic interventions (drug therapy cell-based therapy) disease. summary, review provides systematic elaboration thus shedding light its future research.

Gastrointestinal cancers including esophageal squamous cell carcinoma (ESCC), gastric cancer (GC), and colorectal (CRC) are common highly lethal types of worldwide. Metabolic reprogramming plays a critical role in progression involves metabolic processes such as glucose lipid metabolism. Fatty acid oxidation (FAO) has profound impact on cancer, with many genes cytokines influencing initiation, development, metastasis, resistance by regulating FAO. Additionally, FAO further promotes affecting the tumor microenvironment (TME). The gastrointestinal garnered increasing attention, related anticancer drugs currently being developed.

Succinylation modification involves in the progression of human cancers. The present study aimed to investigate role CPT1A, which is a succinyltransferase prostate cancer (PCa). CCK-8 was used detect cell viability. Seahorse performed evaluate glycolysis. Luciferase assay transcriptional regulation. ChIP assess binding between factors with promoters. Co-IP proteins. We found that CPT1A highly expressed PCa tissues and lines. Silencing inhibited viability glycolysis cells. Mechanistically, promoted succinylation SP5, strengthened SP5 promoter PDPK1. activated PDPK1 transcription AKT/mTOR signal pathway. These findings might provide novel targets for diagnosis or therapy cancer.

The ubiquitin-proteasome system (UPS) is one of the main degradation systems within cells, catalyzing tagging proteins for by ubiquitin molecules, which are then recognized and degraded proteasome. Lipid metabolism plays a crucial role in cellular energy closely associated with occurrence development cancers. Recent research indicates that cancer lipid regulated intracellular proteins, including ubiquitination modifications. This review will explore regulating metabolism, summarize latest progress, propose potential therapeutic strategies.
