Abstract Road dust resuspension in urban environments can contribute to high human exposure metal(loid)s, polycyclic aromatic hydrocarbons, and other potentially toxic organic compounds. However, for many regions, information on loadings, emission factors chemical profiles is lacking accurately apply inventories source apportionment models. People spend most of their time indoors, particularly in houses where daily activities are carried out, enhancing particulate matter (PM) emissions with consequent adverse health impacts. This study intended to appraise the toxicological and mutagenic responses a diameter less than 10 μm (PM10) released from cooking ironing under different conditions. Atmospheric particulate matter (PM) with diameters below 10 µm (PM10) may enter the lungs through inhalation and are linked to various negative health consequences. Emergent evidence emphasizes significance of cell metabolism as a sensitive target PM exposure. However, current understanding relationship between composition, conventional toxicity measures, rewiring intracellular metabolic processes remains limited. In this work, PM10 sampled at residential area (urban background, UB) traffic-impacted location (roadside, RS) Portuguese city was comprehensively characterized in terms polycyclic aromatic hydrocarbons plasticizers. Epithelial lung cells (A549) were then exposed for 72 h organic extracts different biological outcomes assessed. UB RS dose-dependently decreased viability, induced reactive oxygen species (ROS), mitochondrial membrane potential, caused cycle arrest G0/G1 phase, modulated profile. Interestingly, sample, richer particularly toxic PAHs plasticizers, had greater impact than extract. Changes comprised significant increases glutathione, reflecting activation antioxidant defences counterbalance ROS production, together lactate, NAD+, ATP, which suggest stimulation glycolytic energy possibly compensate reduced activity. Furthermore, number other variations hinted changes turnover TCA dynamics, represent novel clues on potential effects.
