This study evaluates the cytotoxic and genotoxic effects of PM2.5 collected from an open-cast coal mining area in northern Colombia. Cyclohexane (CH), dichloromethane (DCM), acetone (ACE) extracts were obtained using Soxhlet extraction to isolate compounds different polarities. Human lymphocytes exposed extracted compounds, cytotoxicity genotoxicity assessed cytokinesis block micronucleus (CBMN) comet assays, incorporating FPG ENDO III enzymes detect oxidative DNA damage. Chemical analysis revealed that organic fractions consisted mainly modified hydrocarbons volatile compounds. The CBMN assay showed a significant increase micronuclei binucleated (MNBN) mononucleated (MNMONO) cells nucleoplasmic bridges (NPB) lymphocytes. substantial damage, particularly with ACE extract, which significantly increased oxidized purines pyrimidines. DCM induced similar effects, while CH moderate effects. CREST immunostaining aneugenic activity, extracts. These results suggest polar PM2.5, likely containing metals PAHs, contribute damage chromosomal instability. highlights need monitor composition regions implement stricter environmental policies reduce exposure health risks.
