Structural and functional alterations in brain microvascular endothelial cells (BMECs) caused by oxygen-glucose deprivation (OGD) are involved the pathogenesis of various disorders. AlkB homolog 5 (ALKBH5) is a primary m6A demethylase that regulates cell processes, but its distinct roles BMEC function remain to be clarified. In present study, mouse middle cerebral artery occlusion (MCAO) model, knockout ALKBH5 reduced neurological deficits, infarct volumes tissue apoptosis ischemia/reperfusion injury. Evans blue leakage decreased expression tight junction protein ZO-1 Occludin were also attenuated knockout. During exploration underlying mechanisms role BMECs, we found was induced at both mRNA levels hypoxia; however, stability impaired OGD treatment.
