Our environment, encompassing air, water, and soil, is replete with a multitude of chemical substances. These chemicals, often derived from everyday manufactured products, such as agrochemicals, pharmaceutical drugs, personal care play significant role in human animal welfare across various sectors. substances can exist individually or mixtures, combining other xenobiotic compounds due to environmental interactions reactions. Plant-parasitic nematodes and insect pests critically threaten agricultural productivity, but chemical pesticides face limitations due to resistance environmental concerns, necessitating eco-friendly biopesticides targeting both nematodes. Here, we developed a high-throughput screening platform using Caenorhabditis elegans identify entomopathogenic fungi exhibiting nematocidal activity against Meloidogyne incognita. Among 32 tested strains, nine Metarhizium spp. one Beauveria strain demonstrated dual efficacy C. M. anisopliae CQMa421 showed the highest virulence, suppressing nematode reproduction by 42.7% inducing >80% mortality. Pot experiments revealed 50% reduction in root galling index 50.3% fewer galls Solanum lycopersicum. The filtrate caused irreversible locomotor deficits reduced egg hatching rates 28%. Concurrently, intestinal damage, elevated oxidative stress autophagy were observed elegans. This was accompanied transcriptome-wide modulation of genes involved detoxification immune defense pathways. These findings demonstrate our elegans-based method for identifying with potential. CQMa421's virulence incognita suggests its promise pest management, while molecular insights highlight pathways that may contribute future design nematicides. study advances fungal biocontrol agents offers sustainable strategy agriculture.
