Extracellular polymeric substances (EPSs) in waste activated sludge (WAS) are important guardians of active microorganisms against toxic emerging contaminants (ECs). However, how ECs interact with EPSs and further interfere microbial functions during WAS anaerobic digestion has not been systemically disclosed. This work reported the dose-dependent effects polyhexamethylene guanidine (PHMG, as a potential EC) on methane production comprehensively revealed underlying mechanisms. PHMG at low dose showed an evident lag period for (from 0.09 d control to 8.23 d) but produced dramatic inhibition high dose. Although promoted solubilization, hydrolysis, acidification via enriching hydrolytic acidogenic bacteria (i.e., Acidaminobacter Acidibacter), methanogenesis was inhibited due reduction methanogens Methanosaeta Syntrophomonas) methanogenic activities related acetoclastic hydrogenotrophic yhdR, pdhB, fwdB, frhD). Meanwhile, interspecies electron transfer between acidogens decreasing biosynthesis shuttles e-pili ribA, yigB, ribBA, MtrD) reducing mutualistic Desulfobulbus Methanospirillum), which resulted decrease formation. Further analysis found that disrupted (mainly proteins) improved cell permeability. Then, it might enter cells induce formation septal invaginations disturb replication well metabolic involved biosynthesis. functional could adapt stress significantly recover upregulating genes growth (e.g., FEN1 ftsQ) activating quorum sensing system (LuxR/I system).
