Hepatocellular carcinoma (HCC) is one of the most prevalent cancers, characterized by high morbidity and mortality rates. Recently, immunotherapy has emerged as a crucial treatment modality for HCC, following surgery, locoregional therapies, targeted therapies. This approach harnesses body's immune system to target eliminate cancer cells, potentially resulting in durable antitumor responses. However, acquired resistance tumor immunosuppressive microenvironment (TIME) significantly hinder its clinical application. advancements nanotechnology, coupled with deeper understanding biology nano-biological interactions, have led development various nanoparticles aimed at enhancing therapeutic efficacy through specific targeting tissues. These increase accumulation immunotherapeutic drugs within microenvironment, thereby transforming TIME. In this review, we provide concise overview fundamental principles governing TIME landscape HCC discuss rationale applications context. Additionally, highlight existing challenges potential opportunities translation nanomedicines.

The development of novel ladder-type conjugated molecules is crucial for advancing supramolecular chemistry and material science. In this study, we report a straightforward synthesis new alternating donor-acceptor (D-A) heteroarene, FCDTDPP, demonstrate its application as photothermal agent imaging cancer therapy. FCDTDPP constructed by vinylene bridge between cyclopentadithiophene (D) diketopyrrolopyrrole (A) through intramolecular Friedel-Crafts type reaction. exhibits unique combination good molecular planarity, efficient intra-/intermolecular mixed D-A interactions, local aromaticity. These features collectively contribute to broad intense absorptions with narrow band gap in red the spectra, coupled multiple vibrational absorption feature, thereby enhancing non-radiative decay process resulting conversion property. nanoparticles (NPs) exhibit superior performance stability under 660 nm laser irradiation. Moreover, vitro studies reveal that NPs possess excellent biocompatibility, low cytotoxicity, robust therapeutic efficacy, finding further corroborated preliminary vivo experiments tumor-bearing mice. This work charts course engineering organic systems, propelling relevant research forward.

Single-wavelength lasers that trigger intelligently designed multifunctional theranostic nanoplatforms are urgently needed for early cancer diagnosis and imaging-guided therapy. In this study, a novel zinc porphyrin, Por-TR, was fabricated by incorporating thiophene as donor introducing electron acceptors on both sides to expand the conjugation. The presence of multiple flexible chains in molecular structure Por-TR inhibits π-π stacking, which allows it form J nanoaggregates when coassembled with DSPE-PEG2000, demonstrating maximum absorption at approximately 808 nm. These NPs provide NIR-II fluorescence/PA dual-modal signals imaging serve combined PTT/PDT therapeutic agent, making them suitable nanoplatform. To further improve their effects, we embedded thermosensitive NO donor, BNN6, nanosystem achieve PDT/PTT/NO Intravenous injection Por-TR-NO into 4T1 tumor-bearing mice enabled accurate observation tumor location via imaging. vivo therapy results show exhibited remarkable antitumor efficacy PTT/PDT/NO therapy, triggered an nm laser. Overall, nanoplatform offers versatile approach treatment.
