Abstract NADPH oxidase 2 (Nox2), a superoxide-generating enzyme, is source of reactive oxygen species (ROS) that regulate the intracellular redox state, self-renewal, and fate hematopoietic stem/progenitor cells (HSPCs). Nox2 complex expressed on HSPCs associated with several activated cell membrane receptors increases level ROS. In addition, ROS are also released from mitochondria and, all together, potent activators pattern recognition receptor Nlrp3 inflammasome, which regulates trafficking, proliferation, metabolism HSPCs. current study, we noticed Nox2-deficient mice, despite increased number in bone marrow (BM), show defects illustrated by delayed recovery peripheral blood (PB) parameters after sublethal irradiation mobilize fewer administration G-CSF AMD3100. Moreover, engraft poorly transplantation into normal syngeneic recipients. To explain these at molecular level, hypothesized Nox2-KO decreased does not efficiently activate plays crucial role regulating trafficking Herein, report display i) defective migration to major chemoattractant, ii) impaired activation iii) defect lipid raft (MLRs) formation required for proper chemotactic response pro-migratory factors. We conclude Nox2-derived enhances inflammasome-dependent manner facilitating MLRs assemble outer membranes, expression results affects migration. Graphical

Abstract Changes in cell state are often accompanied by altered metabolic demands, and homeostasis depends on cells adapting to their changing needs. One major change is senescence, which associated with dramatic changes metabolism, including increases lipid but how accommodate such alterations poorly understood. Here, we show that the transcription factor p53 enables recycling of headgroups required meet increased demand for membrane phospholipids during senescence. activation supply phosphoethanolamine (PEtn), an intermediate Kennedy pathway de novo synthesis phosphatidylethanolamine (PE), transactivating genes involved autophagy lysosomal catabolism enable turnover. Disruption PEtn conversion PE well-tolerated absence results organelle remodeling perturbs growth gene expression following activation. Consistently, CRISPR-Cas9-based genetic screens reveal p53-activated preferentially depend metabolism. Together, these headgroup be a homeostatic function confers cell-state specific vulnerability.

Neural stem cells (NSCs) persist in the adult mammalian brain and are able to give rise new neurons glia throughout life. The largest cell niche mouse is ventricular-subventricular zone (V-SVZ) lining lateral ventricles. Adult NSCs V-SVZ coexist quiescent actively proliferating states, they exhibit a regionalized molecular identity. importance of such spatial diversity just emerging, as depending on their position within niche, distinct subtypes olfactory bulb interneurons different types glia. However, functional relevance heterogeneity still poorly understood. Here, we put into perspective findings highlighting NSC for plasticity, how body signals physiological states regulate behavior.
