analyzed.

Natural
plant-derived
active
ingredients
have
gained
increasing
attention
for
their
potential
in
the
treatment
of
depression
due
to
safety
and
multi-target
pharmacological
activities.
Saikosaponin
A
(SSA),
a
major
bioactive
saponin
extracted
from
Bupleurum
(a
medicine
food
homologous
plant),
has
been
reported
possess
anti-inflammatory,
neuroprotective,
antioxidative
properties.
In
this
study,
we
evaluated
antidepressant-like
effects
SSA
mouse
model
chronic
unpredictable
mild
stress
(CUMS)-induced
depression.
Mice
were
subjected
CUMS,
followed
by
daily
administration
(20
mg/kg,
po
6
weeks).
Behavioral
tests,
including
tail
suspension
test,
open
field
elevated
plus
maze,
marble
burying
indicated
that
significantly
alleviated
depressive-like
anxiety-like
behaviors.
Histopathological
analysis
H&E
staining
showed
reduced
hippocampal
neuronal
damage
induced
stress.
Biochemical
assays
revealed
normalized
levels
neurotransmitters
(5-HT,
DA,
5-HIAA),
enhanced
antioxidant
enzyme
activity
(SOD,
CAT,
GSH),
suppressed
neuroinflammatory
cytokine
production
(TNF-α,
IL-1β,
IL-6).
Mechanistically,
exerted
its
antidepressant
inhibiting
TLR4/NF-κB
signaling
pathway
upregulating
BDNF
expression.
PC12
cells,
TLR4
overexpression
attenuated
SSA’s
protective
effects,
whereas
silencing
cellular
resistance
corticosterone-induced
damage.
These
findings
suggest
alleviates
CUMS-induced
depression-like
behaviors
mice
modulating
neuroinflammation
oxidative
through
TLR4/NF-κB/BDNF
axis,
indicating
as
functional
food-derived

Chemotherapy
(CT)
is

Daphnetin modulates GLP-1R to alleviate cognitive dysfunction in diabetes: implications for inflammation and oxidative stress

Daphnetin
exerts
certain
pharmacological
function
on
a
variety
of
diseases,
but
its
role
in
diabetic
cognitive
dysfunction
has
not
been
elucidated.
In
this
study,
we
carried
series
studies
GLP-1R
with
daphnetin.
rats
and
PC12
cells,
found
that
daphnetin
could
alleviate
increase
the
expression
level
GLP-1R.
Additionally,
anti-diabetic
effect
DAP
was
accompanied
by
inhibition
