Monitoring the physiological changes of organelles is essential for understanding local biological information cells and improving diagnosis therapy diseases. In clinical diagnosis, the level of biological enzymes in serum has been generally regarded as markers human diseases. this work, a kind simple and sensitive plasmonic probe (indicated Au@FeOOH) synthesized with guidance imaging subsequently developed for alkaline phosphatase (ALP) detection under dark-field microscopy (DFM). As hydrolysis enzyme, ALP can promote l-ascorbic acid 2-phosphate to ascorbic (AA). AA further acts strong reduction reagent decomposition FeOOH shell, which results blue shift localized surface plasmon resonance spectra an obvious color change DFM. RGB analyses show that using ΔR/G value instead scattering wavelength or R/G analytical signal, deviation attributed size distribution initial Au NPs is greatly suppressed, linear range from 0.2 6.0 U/L (R2 = 0.99) limit 0.06 are acquired various concentrations during detection. Abstract Alkaline phosphatase (ALP) is an important marker for many diseases, yet efficient and reliable detection method still lacking. Here, we present a novel photonic alginate hydrogel particle as ALP sensor based on the competitive combination of intermediary agent, pyrophosphate ion (PPi), with crosslinker, which can trigger phase-transition response from hydrogel. This be converted into dual optical readouts, fluorescence signal structural color signal. Quantitative determination was established this dual-indicator system high accuracy reliability. These features suggest potential ideal applications responsive particles to detect other macromolecules.
