Bladder cancer (BC) currently ranks as the 9th most common worldwide. It is characterised by very high rates of recurrence and metastasis. Most cases BC are urothelial origin, due to its ability penetrate muscle tissue, divided into non-muscle-invasive (NMIBC) muscle-invasive (MIBC). The current diagnosis still based primarily on invasive cystoscopy, which an expensive method that carries a risk various complications. Tumor hypoxia reshapes the microenvironment, driving progression, invasion, metastasis, and therapy resistance. Patient-derived tumor organoids, formed under three-dimensional conditions, preserve cellular heterogeneity nutrient gradients, making them ideal for studying hypoxia-induced responses. This study examines changes in lung organoids from two patients, focusing on tumor-associated markers, stem cell migration capabilities. Our findings demonstrate that distinctively modulates expression of cancer markers thyroid transcription factor-1, cytokeratin 7, ΔNP63 variant. Hypoxia also induces upregulation cell-associated resulting an increased proportion cells. Furthermore, hypoxic exhibit unique migratory behavior upon reoxygenation, driven by epithelial-mesenchymal transition elevated matrix metalloproteinases 7 9, indicating their enhanced invasive potential.
