Coptidis Rhizoma (CR), a traditional Chinese medicine, has extensive pharmacological activity because it is rich in isoquinoline alkaloids. Nonetheless, the limited solubility of these alkaloids presents significant challenge, hindering full realization CR's therapeutic potential through conventional extraction methods. To address this issue and enhance target compounds while optimizing efficiency, study employed an environmentally sustainable technique, Ultrasound-Enzyme-Trehalose Lipid (UET), to synergistically extract five key from CR. Based on results single-factor experiments, Box-Behnken design (BBD) was optimize selected model variables. The optimal conditions were determined: pH 4.3, temperature 40 ℃, solid-liquid ratio 1:27 g/mL, ultrasonication time 48 min. UET extracts compared with other extracts, proved that had higher alkaloid rate, comprehensive evaluation value (CEV) lower energy consumption CO2 emission. Then scanning electron microscopy (SEM), dynamic light scattering (DLS) transmission (TEM) used explore mechanism extraction. In addition, more excellent anti-inflammatory activity. establishment method CR provides reference for green efficient components natural drugs.

Abstract Coix seed is recognized as a functional medicinal food due to its valuable biological activities, with polysaccharides being the primary active compounds. In this study, an ultrasonic‐assisted enzymatic extraction technique was employed, and response surface methodology used optimize yield of 9.55 ± 0.26%. A novel neutral polysaccharide, CSPsN‐1, purified molecular weight 7.75 kDa. CSPsN‐1 composed arabinose, galactose, glucose, xylose, mannose in molar ratios 0.48: 7.92: 86.39: 2.42: 2.79. Its backbone →4)‐α‐D‐Glc p ‐(1→ →3,4)‐α‐D‐Glc units, terminal residues α‐D‐Glc . vitro experiments, enhanced glucose consumption insulin‐resistant HepG2 cells upregulated GLUT4 expression by activating PI3K/AKT signaling pathway. These findings suggest that holds significant promise ingredient for treating insulin resistance related metabolic disorders.
