Oral squamous cell carcinoma (OSCC) is a common malignant tumor in the head and neck, associated with high recurrence poor prognosis. We performed an integrated analysis of single-cell RNA spatial transcriptomic data from cancerous normal tissues to create comprehensive atlas epithelial cells cancer-associated fibroblasts (CAFs). Objective Hepatocellular carcinoma (HCC) poses a significant clinical challenge because the long-term benefits of immune checkpoint blockade therapy are limited. A comprehensive understanding mechanisms underlying immunotherapy resistance in HCC is imperative for improving patient prognosis. Design In this study, to systematically investigate characteristics cancer-associated fibroblast (CAF) subsets and dynamic communication among tumor microenvironment (TME) components regulated by CAF subsets, we generated an atlas compiling single-cell RNA sequencing (scRNA-seq) datasets on 220 samples from six datasets. We combined spatial transcriptomics with scRNA-seq multiplexed immunofluorescence identify specific TME that determine efficacy patients. Introduction: Among the various stromal cell types within tumor microenvironment, cancer-associated fibroblasts (CAFs) emerge as predominant constituent, exhibiting a diverse array of oncogenic functions not intrinsic to normal fibroblasts. Their involvement spans across all stages tumorigenesis, encompassing initiation, progression, and metastasis. Current understanding posits coexistence distinct subpopulations CAFs microenvironment spectrum solid tumors, showcasing both pro- antitumor activities. Recent advancements in single-cell transcriptomics have revolutionized our ability meticulously dissect heterogeneity inherent CAF populations. Furthermore, accumulating evidence underscores pivotal role conferring therapeutic resistance tumors against drug modalities. Consequently, efforts are underway develop pharmacological agents specifically targeting CAFs.
