According to the World Health Organisation and European Commission definitions, substances shall be considered as having endocrine disrupting properties if they show adverse effects, have activity effects are a consequence of (using weight-of-evidence approach based on biological plausibility), unless not relevant humans or non-target organisms at (sub)population level. To date, there is no decision logic how establish disruption via thyroid modality in non-mammalian vertebrates. This paper describes an evidence-based compliant with integrated testing assessment (IATA) concept, identify thyroid-mediated effect patterns aquatic vertebrates using amphibians models for assessment. The includes existing test guidelines methods proposes detailed considerations select assays interpret findings. If mammalian dataset used starting point indicates concern, Xenopus Eleutheroembryonic Thyroid Assay allows ruling out vertebrates, whereas Amphibian Metamorphosis its extended, fixed termination stage variant inform both potentially population-relevant adversity. In evaluating findings, response all assay endpoints considered, including direction changes. Thyroid-mediated identified individual level amphibian tests followed by mode-of-action population relevance assessments. Finally, data overarching evaluation. has been designed generically can adapted, e.g. accommodate fish once available It also ensures that scientifically information animal minimised. proposed included regulatory assessments facilitate conclusion whether meet disruptor definition

ABSTRACT Fish in the eleutheroembryonic life stage are defined as embryos or hatched fry before external self‐feeding begins, and this is not classified a protected according to EU (Directive 2010/63/EU) because of its alignment with 3R principles (replacement, reduction, refinement). In Minami‐medaka ( Oryzias latipes ), considered extend until hatching, OECD TG210, whereas no supporting evidence identify has yet been reported. To clarify medaka stage, we investigated effects non‐feeding on survival, growth, gene expression NIES‐R, Hd‐rR, d‐rR strains. Non‐feeding did affect survival up 6 days post‐hatching (dph) any strain, rates exceeding 80%. However, beyond 8 dph reduced below 50% at 30 dph. measured total length, was significantly affected by dph, except for Hd‐rR. Analysis differentially expressed genes (DEGs) response revealed that autophagy‐related DEGs wipi2 , wdr45 wipi1 atg14 map1lc3b ) were found from 43 genes. other upregulated after 4 non‐feeding, respectively. effect rescued feeding. Together, < be least suggesting can used evaluate chemical toxicity endocrine‐disrupting activity principles. This study indicated strains Minami‐medaka, fish growth Hd‐rR strain.
