Multiple sclerosis is an autoimmune disease with a strong neuroinflammatory component that contributes to severe demyelination, neurodegeneration and lesions formation in white grey matter of the spinal cord brain. Increasing attention being paid signaling biogenic amine histamine context several pathological conditions. In multiple sclerosis, regulates differentiation oligodendrocyte precursors, reduces improves remyelination process. However, concomitant activation H1-H4 receptors can sustain either damaging or favorable effects, depending on specifically activated receptor subtype/s, timing engagement, central versus peripheral target district. Conventional drug development has failed so far identify curative drugs for thus causing delay therapeutic options available patients. this perspective, repurposing offers exciting complementary alternative rapidly approving some medicines already approved other indications. present work, we have adopted new network-medicine-based algorithm called SAveRUNNER, quantifying interplay between sclerosis-associated genes targets human interactome. We identified drug-disease associations predicted off-label novel use histaminergic amodiaquine, rupatadine, diphenhydramine among others, sclerosis. Our work suggests selected histamine-related molecules might get root causes emerge as potential strategies disease.

Hyperglycemia, and exacerbation of pre-existing deficits in glucose metabolism, are manifestations the post-acute sequelae SARS-CoV-2. Our understanding metabolic decline after acute COVID-19 remains unclear due to lack animal models. Here, we report a non-human primate model SARS-CoV-2 using infected African green monkeys. Using this model, identify dysregulated blood chemokine signature during that correlates with elevated persistent hyperglycemia four months post-infection. Hyperglycemia also liver glycogen levels, but there is no evidence substantial long-term replication pancreas. Finally, favorable glycemic effect mRNA vaccine, administered on day 4 Together, these data suggest monkey exhibits important similarities humans can be utilized assess therapeutic candidates combat COVID-related defects.

When acute SARS-CoV-2 infections cause symptoms that persist longer than 3 months, this condition is termed long COVID. Symptoms experienced by patients often include myalgia, fatigue, brain fog, cognitive impairments, and post-exertional malaise (PEM), which the worsening of following mental or physical exertion. There little consensus on pathophysiology exercise-induced PEM skeletal-muscle-related symptoms. In opinion article we highlight intrinsic mitochondrial dysfunction, endothelial abnormalities, a muscle fiber type shift towards more glycolytic phenotype as main contributors to reduced exercise capacity in The mechanistic trigger for induce unknown, but rapid skeletal tissue damage intramuscular infiltration immune cells contribute PEM-related

Abstract Sarbecoviruses such as SARS and SARS-CoV-2 have been responsible for two major outbreaks in humans, the latter resulting a global pandemic. While sarbecoviruses primarily cause an acute respiratory infection, they shown to infect nervous system. However, mechanisms of sarbecovirus neuroinvasion neuropathogenesis remain unclear. In this study, we examined infectivity trans-synaptic transmission potential human stem cell–derived neural model systems. We demonstrated limited ability replicate neurons. Furthermore, inability transmit between synaptically connected Finally, determined absence infection olfactory neurons experimentally infected ferrets. Objectives This study aims to characterise the diversity of post-COVID-19 physical and mental health outcomes, known as condition (PCC), determining factors 3–6 months after acute SARS-CoV-2 infection. Design is a prospective cohort study. Setting took place at European Hospital Marseille, France. Participants include patients with COVID-19 treated inpatients or outpatients. Interventions face-to-face assessment symptoms. Main outcome measures symptom scores scales, well paraclinical elements (thoracic CT scan, pulmonary functional tests). Multiple component analysis was used identify clinical phenotypic clusters PCC patients, their initial comorbidity groups. A multinomial regression model evaluate association between comorbidities disease severity phenotype. Results total 210 agreed participate, which 157 (75%) reported least one visit; mostly asthenia, dyspnoea, psychiatric disorders such anxiety, depression, post-traumatic stress disorder cognitive disorders. Four were recognised: (1) paucisymptomatic (n=82, 39%); (2) sequelae (n=39, 18.6%), (3) pre-existing (n=29, 13.8%); (4) somatic and/or symptoms (n=60, 28.6%). In addition symptoms, in these differed terms demographic characteristics (sex), COVID-19. Conclusions The four identified corresponded distinct coherent entities, making it possible consider adapted personalised prognosis therapeutic interventions.

Long COVID has emerged as a significant challenge since the COVID-19 pandemic, which was declared an outbreak in March 2020, marked by diverse symptoms and prolonged duration of disease. Defined WHO persisting or emerging for at least two months post-SARS-CoV-2 infection without alternative cause, its prevalence varies globally, with estimates 10–20% Europe, 7.3% USA, 3.0% UK. The condition’s etiology remains unclear, involving factors, such renin–angiotensin system overactivation, persistent viral reservoirs, immune dysregulation, autoantibodies. Reactivated viruses, like EBV HSV-6, alongside epigenetic alterations, exacerbate mitochondrial dysfunction energy imbalance. Emerging evidence links SARS-CoV-2 to chromatin gut microbiome changes, further influencing long-term health impacts. Diagnosis long requires detailed systemic evaluation through medical history physical examination. Management is highly individualized, focusing mainly on patient’s affected systems. A multidisciplinary approach essential, integrating perspectives address manifestations, underlying mechanisms, therapeutic strategies. Enhanced understanding COVID’s pathophysiology clinical features critical improving patient outcomes quality life. With growing number cases expected advancing research disseminating knowledge remain vital developing effective diagnostic management frameworks, ultimately supporting better care individuals.
