Herein, we proposed a versatile G-quadruplex (G4)-tetrahedral DNA framework (G4-TDF) nanostructure functionalized origami microfluidic paper-based device (μPADs) for fluorescence detection of K+ by lighting up thioflavin T (ThT). In this work, TDF provided robust structural support G-rich sequence in well-defined orientation and spacing to ensure high recognition efficiency, enabling sensitive sensing on μPAD. After introducing ThT, the sequences extended from vertices formed parallel G4 structure, showing weak signal output. Upon presence target K+, structure transitioned antiparallel leading significantly increase ThT. Benefiting outstanding enhancement characteristic ThT excellent specificity plus satisfactory efficiency with aid TDF, strategy exhibited an impressive limit as low 0.2 mM wide range 0.5–5.5 mM. This innovative G4-TDF was applied first time μPAD, providing simple, effective, rapid method human serum significant potential clinical diagnostics.

Developing powerful immunoassays for sensitive and real-time detection of targets has always been a challenging task. Due to their advantages direct readout, controllable sensing, low background interference, photothermal have become type new technology that can be used various applications such as disease diagnosis, environmental monitoring, food safety. By modification with antibodies, materials induce temperature changes by converting light energy into heat, thereby reporting specific target recognition events. This article reviews the design application based on different materials, including noble metal nanomaterials, carbon-based two-dimensional oxide sulfide Prussian blue nanoparticles, small organic molecules, polymers, etc. It pays special attention role working principle immunoassays. Additionally, challenges prospects future development are briefly discussed.
