Plastic particles impact the propagation of antibiotic resistance genes (ARGs) in environmental media, and their perturbation on horizontal gene transfer (HGT) ARGs is recognized as a critical influencing mechanism. However, studies concerning influence mechanisms plastic HGT were limited, particularly for effect particle sizes ARG vector-associated mechanisms. Microplastics (MPs) as emerging contaminants have become a global environmental problem. However, studies on the effects of MPs metabolic diseases remain limited. Here, we evaluated polystyrene (PS), one most prominent types MPs, insulin sensitivity in mice fed with normal chow diet (NCD) or high-fat (HFD), and explained underlying mechanisms. Mice NCD HFD both showed resistance (IR) after PS exposure accompanied by increased plasma lipopolysaccharide pro-inflammatory cytokines such tumor necrosis factor-α interleukin-1β. Exposure to also resulted significant decrease richness diversity gut microbiota, particularly an increase relative abundance Gram-negative bacteria Prevotellaceae Enterobacteriaceae. Additionally, small particle size (5 μm) accumulated liver, kidneys blood vessels mice. Further analyses inhibition signaling pathway liver exposed mice, IRS1 decreased expression PI3K. Hence, mechanism induce IR might be mediated through regulating microbiota accumulation tissues, stimulating inflammation inhibiting pathway. In conclusion, potential contaminant that causes associated IR.
