Polybrominated diphenyl ethers (PBDEs) are persistent organic pollutants prevalent in the environment. Organohalide-respiring bacteria (OHRB) can attenuate PBDEs via reductive debromination, but often producing toxic end-products. Debromination of to ether remains a rare phenomenon and is so far specifically associated with Dehalococcoides isolated from e-waste polluted sites. The occurrence PBDE debromination other ecosystems underpinning OHRB underexplored. Here we found that common trait sewage sludge microbiota, was produced as end-product at varying quantities (0.6-52.9% mol parent PBDEs) 76 84 cultures established bioreactor sludge. Diverse pathways converting ether, including several new routes, were identified. Although contributed Dehalogenimonas, Dehalobacter, uncultivated Dehalococcoidia likely played more important roles than previously recognized. Multiple dehalogenase genes (including bdeA, pcbA4, pteA, tceA) also coexisted Collectively, these findings contribute enhancing our comprehension environmental fate PBDEs, expanding diversity microorganisms catalyzing developing consortia for bioremediation application.

Polychlorinated biphenyls (PCBs) are pervasive pollutants that pose risks to ecosystems and human health. Microbial reductive dehalogenation plays crucial roles in attenuating PCBs, but comprehensive insights into PCB dechlorination pathways, reactivity, governing factors limited by the vast number of congeners costly experimental approaches. We address this challenge establishing a high-throughput vitro assay approach (HINVARD), which increases test throughput 30-fold enhances reagents cell utilization efficiency over 10-fold compared conventional methods. Using HINVARD, we screened 61 across 9 enrichment cultures 3 Dehalococcoides isolates, identifying active 31-44 congeners. Results showed congener properties (chlorine substitution patterns, steric hindrance, solubility) primarily determine potential, leading consistent reactivity trends cultures. In contrast, different organohalide-respiring bacteria catalyzed distinct preferentially removing para- or meta-chlorines. Structural modeling dehalogenases revealed unique binding orientations substrate specificity, offering molecular these pathways. This study provides high-efficiency strategy for investigating microbial dehalogenation, yielding first understanding patterns mechanisms. These findings guide design tailored consortia effective bioremediation.

Halogenated organic compounds are persistent pollutants that pose a serious threat to human health and the safety of ecosystems. Cobamides essential cofactors for reductive dehalogenases (RDase) in organohalide-respiring bacteria (OHRB), which catalyze dehalogenation process. This review systematically summarizes impact cobamides on organohalide respiration. The catalytic processes cobamide also discussed.
