Insulin-like peptides (ILPs) are important peptide hormones in insects, particularly involved regulating physiological processes such as growth, development, and reproduction. However, the specific roles of ILPs reproduction natural enemy insects remain unknown. In this study, two ILP genes, CsILP1 CsILP2, were cloned their functions analyzed female Coccinella septempunctata L. (Coleoptera: Coccinellidae). The open reading frames (ORFs) CsILP2 384 bp 357 bp, respectively. expression increased on 6th day after eclosion, reaching its peak 12th day, while levels showed a significant increase then stabilized. different tissues, was highly expressed ovaries, predominated elytra. Injection dsRNA targeting resulted down-regulation insulin pathway genes. relative ovarian development-related genes Vasa, G2/M, Vg reduced by 82.50%, 89.55%. 96.98% dsCsILP1-treated females, 42.55%, 91.36%, 55.63% dsCsILP2-treated females. Furthermore, substantial decreases 14-day fecundity observed, with reductions 89.99% for dsCsILP1 83.45% dsCsILP2. These results confirm regulatory C.

Energy metabolism is essential for insect development, reproduction and detoxification. Insects often reallocate energy resources to manage external stress, balancing the demands of detoxification reproduction. Glucose transport 4 (Glut4), a glucose transporter, involved in lipid metabolism. However, specific molecular mechanism Glut4 reproduction, its role response insecticide-induced oxidative stress remain unclear. In this study, LmGlut4 was identified analyzed Locusta migratoria. Silencing significantly reduced vitellogenin (Vg) biosynthesis fat body Vg absorption by oocytes, ultimately hindering ovarian development oocyte maturation. Knockdown also inhibited key hormones, such as juvenile hormone (JH), 20-hydroxyecdysone (20E) insulin. Furthermore, knockdown led triglyceride (TG) glycogen content ovary, well decreased capacity trehalose adipocytes. Additionally, dsLmGlut4-treated locusts showed heightened sensitivity deltamethrin, leading increased depletion during This study sheds light on biological function ovary provides potential target genes exploring pest management strategies.

Insulin signalling pathways play crucial roles in regulating growth and development insects, but their effects on the of Arachnids, such as spiders, have rarely been studied. As a valuable pest natural enemy agricultural fields, molecular mechanisms insulin pathway-mediated wolf spider, Pardosa pseudoannulata, are particular interest.
