ABSTRACT Fatty acid binding protein 7 (FABP7) is prominently expressed in astrocytes and a critical regulator of inflammatory responses. Accumulating evidence suggests that FABP7 crucial neuropsychological disease through the modulation spinogenesis. Nonetheless, impact on depressive disorders underlying mechanisms not fully understood. Here, we investigated antidepressant properties using chronic unpredictable mild stress (CUMS)‐induced model depression possible mechanisms. Our results revealed depressive‐like behavior induced by CUMS was associated with decreased levels hippocampus (HP). Furthermore, overexpression HP mitigated increased expression its downstream target caveolin‐1 (Cav‐1). specifically regulates astrocyte marker GFAP, as well blood–brain barrier (BBB)‐associated proteins AQP4, CLDN‐5, occludin, LRP1. Notably, CUMS‐induced upregulation pro‐inflammatory factors IL‐1β IL‐6 also significantly reversed HP. This intervention led to postsynaptic proteins, including PSD95 GluA1, an increase brain‐derived neurotrophic factor (BDNF) enhanced neuronal dendritic spine density. The findings indicate exerts antidepressant‐like inhibiting inflammation, regulating spinogenesis, modulating BBB‐related proteins.

As the global elderly population is rising, concerns about cognitive decline and memory loss are becoming urgent. This study evaluated potential of sea cucumber hydrolysates (SCH) from Stichopus japonicus in alleviating deficits using a D-galactose-induced murine aging model. The effects SCH on behavior, hippocampal morphology, gut microbiota, cholinergic system, brain-derived neurotrophic factor (BDNF) signaling, neuroinflammatory pathways were investigated. Results showed that ameliorated learning reduced neuronal damage mice. also modulated along with increased fecal short-chain fatty acids levels. Functional prediction revealed alterations microbiota related to signal transduction. Further, enhanced function through elevating acetylcholine (ACh) levels inhibiting acetylcholinesterase (AChE) activity activated BDNF consistent predictions function. Restoration homeostasis transmission pathway might contribute inhibition neuroinflammation via suppressing microglial activation nuclear kappa-B (NF-κB) pathway. In summary, attenuated neuroinflammation, which be correlated transduction caused by regulating microbiota. Further validation will conducted depletion transplantation. These findings suggest promising functional component for counteracting aging-related deficits.

Depression is characterized by prominent indicators and manifestations, such as anhedonia, which refers to the inability experience pleasure, persistent feelings of hopelessness. In clinical practice, primary treatment approach involves utilization selective serotonin reuptake inhibitors (SSRIs) related pharmacological interventions. Nevertheless, it crucial recognize that these agents are associated with significant adverse effects. Traditional Chinese medicine (TCM) adopts a multifaceted approach, targeting diverse components, multiple targets, various channels action. The microbiota stability, diversity, and composition are pillars for an efficient beneficial symbiotic relationship between its host itself. Microbial dysbiosis, a condition where homeostatic bacterial community is disturbed by acute or chronic events, predisposition many diseases, including local systemic inflammation that leads to metabolic syndrome, diabetes, some types of cancers. Classical dysbiosis occurs in the large intestine. During this period, pathogenic strains can multiply, taking advantage compromised environment. This overgrowth triggers exaggerated inflammatory response from human immune system due weakened integrity intestinal barrier.
